| Aims: To investigate the expression of CD151and CO-029in intrahepaticcholangiocarcinoma tissue,and to analyze the relationship between the expression ofCD151and CO-029and clinical pathological indexes,in order to provide basis forclinical prevention and treatment of intrahepatic cholangiocarcinoma.Materials and methods: The tissue samples and clinical data were collectedfrom patients in Hunan Provincial People,s Hospital from August2012to January2014,which were underwent surgery. The experiment were divided into three groups;Group A for normal intrahepatic bile ducts (liver trauma resection or liverhemangioma specimens, already pathological detection is no cancerous tissue);Group B for intrahepatic cholangiocarcinoma near bile duct cancer group (fromoutside bile duct carcinoma tissue2cm of cancerous tissue, the pathology inspectionfor not cancerous tissue); Group C for intrahepatic cholangiocarcinoma (confirmed bypathology of cancerous tissue)。A kind of method is: using the method of real-timePCR detection of intrahepatic cholangiocarcinoma tissue, the tissue adjacent tocarcinoma and normal bile duct tissues fresh specimens of CD151and CO-029in theexpression of mRNA level;Another kind of method is: using immunohistochemistry(S-P) method to observe the intrahepatic cholangiocarcinoma tissue, the tissueadjacent to carcinoma and normal bile duct tissue paraffin section of the basicpathological characteristics。By comparing statistical software analysis intrahepaticcholangiocarcinoma tissue expression of CD151and CO-029with intrahepaticcholangiocarcinoma of the relationship between clinical pathological indicators, aswell as CD151and CO-029protein expressed in intrahepatic cholangiocarcinomatissue. Results: Real-time PCR results showed that CD151mRNA in the relativeexpression level in intrahepatic cholangiocarcinoma tissue were significantly higherthan tissue adjacent to carcinoma and normal bile duct tissue (p <0.05). CO-029mRNA expression level relatively same statistical differences among three groups,intrahepatic cholangiocarcinoma tissue relative CO-029mRNA expression levelshould also be significantly higher than tissue adjacent to carcinoma and normal bileduct tissues (p <0.05); Immunohistochemical results show that CD151protein inintrahepatic cholangiocarcinoma tissue, the tissue adjacent to carcinoma and normalbile duct tissues expression differences of CD151protein expression in the tissueadjacent to carcinoma and normal bile duct tissues positive rate is lower than theintrahepatic bile duct carcinoma tissue (p <0.05), intrahepatic cholangiocarcinomatissue of CD151protein expression in different degree of differentiation, differenttumor stage and whether there are differences between patients with lymph nodemetastasis. The higher the lower the degree of differentiation, TNM staging, lymphnode metastasis is CD151protein expression in patients with positive rate is higher.CO-029protein expression positive rate in intrahepatic cholangiocarcinoma tissuethan in tissue adjacent to carcinoma and normal bile duct tissue (p<0.05),intrahepatic cholangiocarcinoma tissue of CO-029protein expression differencesbetween different degree of differentiation of patients. The lower the degree ofdifferentiation, CO-029protein expression positive rate is higher. In CD151proteinpositive expression of intrahepatic cholangiocarcinoma group, CO-029has highexpression, the Pearson correlation analysis, intrahepatic cholangiocarcinoma tissueof CD151protein expression and CO-029protein expression was positivelycorrelation (r=0.4, p <0.01), with statistical significance.Conclusions:1. The CD151protein and CO-029protein is highly expressed in the intrahepaticcholangiocarcinoma tissue.2. CD151protein expression in the tissue of intrahepatic cholangiocarcinoma andpatients with TNM staging, differentiation degree and lymph node metastasis. 3. The CO-029protein in patients with intrahepatic cholangiocarcinoma tissue of theexpression and the degree of differentiation.4. Intrahepatic cholangiocarcinoma tissue expression of CD151protein and CO-029protein exist positive correlation. |
