Objective: To study the effect of total oil in Stahlianthus involucratusongrowth inhibition in CNE-2cancer cell and the action mechanism of drug.Methods: CNE-2cells were the experiment cells. Experimentation weredivided into120μg·mL-1、240μg·mL-1、360μg·mL-1、480μg·mL-1control groupand performed after24h. The proliferations and half inhibitionconcentration(IC50) were observed by MTT assay in CNE-2cells. The hologicalchanges of apoptosis of CNE-2cells were determined by Hoechst333258fluorescent staining. FCM assays were used to detect cell apoptotosis and cellcycle;content of reactive oxygen species (ROS) were inspected by flowcytometry; JC-1dyeing method was used to detect the potential change ofCNE-2cells mitochondrial membrane.expressions of caspase-3、bcl-2wasexamined by semi-quantitate reverse transcript PCR assay.Results: After total oil in Stahlianthus involucratus treatment, theproliferation of human nasopharyngeal cancer line CNE-2was blocked in adose-and time-dependent manner. The IC50values of Stahlianthus involucratus for human nasopharyngeal cancer line CNE-2, was300μg·mL-1by MTT methodon24h. After CNE-2cancer cell was treated by the total oil in Stahlianthusinvolucratus, typical morphological changes of apoptosis were observed. Theapoptotic cell were detected after treating with120μg·mL-1,240μg·mL-1and360μg·mL-1of the total oil in Stahlianthus involucratus for24h and the effectwas enhanced as the dosage increased. Cell cycle was also analyzed, with resultsshowing that total oil in Stahlianthus involucratus leads to obstructing of G1stage.The total oil of Stahlianthus involucratus can increase of the cellular ROSlevel, decrease of△Ψm, the total oil (120μg·mL-1、240μg·mL-1) can increase theexpressions of caspase-3and decrease the expressions of bcl-2.Conclusions: Total oil can effectively inhibit growth of CNE-2cancer cell.Its mechanism of anti-cancer action could be attributed to its action of initiatingthe apoptosis of cancer cell. These results indicated thatthe possible mechanismof total oil-inducing cell apoptosis was associated with imbalance ofoxidation-reduction state, increase of the cellular ROS level, destruction ofmitochondrial function, decrease of△Ψm, and caspase pathway activation. |
