BackgroundAcute mountain sickness(AMS) is excitability reaction of organism to plateauenvironment, it is the Clinical symptoms through the regulation of neurohumour onthe new basic to the new equilibrium. The symptom of AMS include headache,flustered, decreased food appetite, tireness, nausea, deadlimb and tics and so on.AMS is detected in prophase, the symptom can be controlled, if not,the developmentof pathogenetic condition aggratates quickly, leading to encephaledema andpneumonedema even to threaten life. The rate of effluence risk of AMS is closelycorrelated with hypoxia environment and ascending speed[1]. If at the altitude of4000m, ascending speed is91m·h-1,54%of the subjects were AMS; If ascending speedis1268m·h-1, the incidence rate of AMS were73%; If ascending speed is1647m·h-1, the incidence rate of AMS were89%[2].It is very important to interfere in AMS in time, we have no desirable methodand test facility of AMS so far. The aim of this research is to compare the expressionof HIF-1(hypoxia inducing factor1), RelA (nuclear factor-KappaB p65), VEGF(vascular endothelial growth factor) and P38MAPK before ascending of plateau and after ascending of4000m altitude and their relationships with AMS, to provideexperiment base for prevention and cure of AMS.ObjectiveTo explore the change of expression of hypoxia inducing factor1(HIF-1),P38MARK, nuclear factor-KappaB p65(RelA)and vascular endothelial growth factor(VEGF) after ascenting of plateau in vivo, and the correlation with the pathogenesyof AMS.MethodsCollecting soldiers’ venous blood before they asent of plateau and after theyarrive in Yushu in Qinghai province48h later respectively, extractingmononuclearcell the expression of mRNA of HIF-1, RelA and P38MARK weredetected by RT-PCR and the expression of protein of HIF-1and VEGF was detectedby Western blotting. The AMS is diagnosed according to《The Diagnose AndProcessing Principle Of AMS》, and the correlation between the expressions ofHIF-1and RelA and pathogenesis of AMS was analyzed.ResultsAfter ascentting of plateau the mRNA expression levels of HIF-1, RelA,P38MARK and the protein of HIF-1and VEGF all increase significancely(p<0.01).At the same time, the change of expression levels of HIF-1and VEGF protein arehighly corellated with AMS, the coefficient correlation is separately0.806and0.675(p<0.001).ConclusionsThe expression level of HIF-1, RelA, p38MARK and VEGF increasesignificantly after asentting of plateau, especially the changes of HIF-1protein andVEGF, it is highly correlated with the disease severity of AMS. The expression levelsof HIF-1and RelA mRNA are significantly increased48h after ascentting ofplateau,and they have positive correlation, prompting the existence of cross-talk phenomenon between their signal transduction pathways.
