ObjectiveHepatocellular carcinoma (Liver cancer) is a malignant tumors which occurs in the liver, including hepatocellular carcinoma and metastatic liver cancer. Its incidence and mortality is high. So, it is a serious threat to human life and health. The traditional treatment for liver cancer which is also the main treatment is surgical operation, radiotherapy, chemical anti-tumor drug treatment. However, the effect of traditional treatment is not ideal, and the recurrence and metastasis of hepatocellular carcinoma is difficult to control. In recent years, more and more researches have been done for antineoplastic effect of natural plant. In this experiment we extracted and separated daidzein, Mung, Gin and L08from soybean, mung bean, Ganoderma lucidum and ginger, and treated liver cancer SMMC-7721cells in vitro with these four kinds of drugs.So, we can study their effection on the proliferation of hepatocellular carcinoma cell SMMC-7721and the expression of CD44in liver cancer stem cell,and want to provide new experimental data for the prevention and treatment of liver cancer.Methods1. Human liver cancer cell SMMC-7721was cultivated to acquire the cell in exponential phase in vitro.2. Human liver cancer cell SMMC-7721which is that in exponential phase was separately disposed by Daidzein, Mung, L08and Gin which the concentration are100μg/ml,200μg/ml,300μg/ml,400μg/ml and500μg/ml. That times separately are 24h,48h and72h. After that, the rate of cell proliferate was determined by MTT and the curve of cell growth was drawn.3. Human liver cancer cell SMMC7721was disposed by Daidzein, Mung, L08and Gin which the concentration are100μg/ml,300μg/ml and500μg/ml. That time is24h. Antigen was detected on the cell surface by this process that Antibody which is marked by fluorescein and cell were detected by Flow cytometry after they were mixed.4. Human liver cancer cell SMMC-7721was disposed by Daidzein, Mung, L08and Gin which the concentration are100μg/ml,300μg/ml and500pg/ml. And the level of liver cancer stem cells CD44protein expression determined by Western Blot.Results1. Test result of MTT:Daidzein, Mung, L08and Gin are able to inhibit the proliferation of human liver cancer cell SMMC-7721in different concentration and different administration time. There is a closely connection between concentration and effect of inhibition.2. Test result of Flow cytometry:human liver cancer cell SMMC-7721was separately disposed by Daidzein, Mung, L08and Gin more than24h. The number of CD44positive cells which were detected in human liver cancer cells is less than that of control group.3. Test result of Western Blot:The level of CD44protein expression of human liver cancer cells after the treatment by Daidzein, Mung and L08in experimental concentrations is lower, and the level of CD44protein expression is influenced by Dose of Daidzein, Mung and L08. However, Gin is unable to lower the level of CD44protein expression of human liver cancer cell.Conclusions1. Daidzein, Mung, L08and Gin are able to inhibit the proliferation of human liver cancer cell SMMC-7721, and the concentrations influence the effect of inhibition.2. Daidzein, Mung, L08and Gin are able to inhibit the proliferation of CD44positive cell of human liver cancer cell SMMC-7721.3. Daidzein, Mung and L08could lower the level of CD44protein expression of human liver cancer cell. But Gin could not lower the level of CD44protein expression.
