Objective: TCM believes that "lung advocate fur", according to this theory, SSc inpatients with systemic sclerosis (Systemic scleroderma, SSc) and the incidence of lungshould be closely related to. Modern medicine thinks the cases of scleroderma isexcessive extracellular matrix accumulation cause skin and visceral fibrosis, andmatrix metalloproteinases play an important role in matrix degradation. But at thesame time, MMPs (Matrix metalloproteinases, MMPs) and its inhibitor TIMPs (Tissueinhibitor of metalloproteinases TIMPs) regulation, in physiological conditions, MMPsand TIMPs maintained a dynamic equilibrium. If the balance is destroyed it will causeexcessive accumulation of extracellular matrix, which occurred in the skin andvisceral fibrosis. MMP-1, TIMP-1in the process of tissue fibrosis play a major role,therefore, to observe the pulmonary represents Maimendong Decoction on MMPsmouse model of bleomycin induced scleroderma lung and skin (MMP-1) and TIMPs(TIMP-1) effects can be discovered, modern TCM Mechanism "lung advocate fur", atthe same time to investigate the preventive effect of Ophiopogon Decoction on SSc.Ideas and provide the reference for the clinical treatment of scleroderma.Methods:200ug/ml of bleomycin (bleomycin, BLM)0.1ml was injected into thedorsal skin of female BALB/c mice, once a day, for three consecutive weeks, constructthe model of scleroderma, followed by observing the changes of hydroxyprolinecontent of skin, skin damage the skin and lung tissue, the colorimetric determinationof pathological morphology, skin and lung tissue in to verify the success of modeling.After the success of the modeling by random principle mice were divided into blankgroup, model group, Radix Ophiopogonis Decoction high dose group, middle dosegroup, Ophiopogon Decoction Ophiopogon Decoction low dose group and Bozhitablets group, each group of12mice. Medication after three weeks of treatment,collecting specimens, cut the mice back skin and lung area, the skin and lung tissuefixed in neutral formalin solution of10%, MMP,-1were determined byimmunohistochemical method, TIMP-1in the skin and lung tissues. Results:(1) with bleomycin induced scleroderma mice model.(2) Maimendong decoction can improve the high expression of MMP-1modelmice, reduce the low expression of TIMP-1.Conclusion:(1) Maimendong decoction has a good therapeutic effect on SSc modelmice.(2) the mechanism of Ophiopogon Decoction in the treatment of SSc micemay be related to the increase expression of model group mice lung and skin tissue ofMMP-1, reduce the TIMP-1.
