Objective:To study the cell and molecular biology mechanism of Yiqi HuayuQingre prescription and its separations in the treatment of mesangial proliferativeglomerulonephritis(MsPGN) on to the proliferation in mesangial cells in vitrostimulated by lipopolysaccharide(LPS) and the effects of both on the expression ofmRNA and protein of NF-κBp65and IκB-α.Methods:Adopte molecular biology and pharmacology of serum methods tostudy the effect of Yiqi Huayu Qingre prescription and its separations on proliferationin the MsC in vitro and observe the effect of Yiqi Huayu Qingre party and its separateon the expression of NF-κBp65and IκB-αmRNA at cell and molecularbiology level.The rat MsC stimulated by LPS was divided randomly into eight equal groups base onconventional culture:normal group,model group,control group,contained-yiqi serumgroup,cntained-huayu Serum group,contained-qingre serum group,ContainedCompound serum group,contained-Tripterygium wilfordi serum group.Using theWestern assay to detect the effect of expression protein of NF-κBp65and IκB-α;Using RT-PCR method to detect the effect of expression mRNA of NF-κBp65andIκB-α.Results:1.The results showed that all containing medicine serums canrestrained the expression of NF-κBp65and enhanced the expression of IκB-α.Compared with the control group each containing medicine serum hadgroupsignificant difference (P <0.05),indicating a successful model.About theresults of restraining the expression mRNA of NF-κBp65,compared withcontained-compound serum group,the effects of contained-Tripterygium wilfordiserum group and contained-huayu serum group were much stronger (P <0.05),butthe other groups had no significant difference(p>0.05)2..About the results of restraining the expression mRNA of IκB-α,the effect ofcontained-compound serum group was the strongest among all the groups,and contained-yiqi serum group was the weakest(P <0.05).3.About the results of restraining the expression protein of NF-κBp65,Compared with contained-compound serum group,the effects of contained-TripterygiUmwilfordi serum group and contained-huayu serum group were much stronger (P <0.05),but the other groups had no significant difference(p>0.05).4.About the results of restraining the expression protein of IκB-α,the effect ofcontained-compound serum group was the strongest among all the groups,andcontained-yiqi serum group was the weakest(P <0.05).Conclusion: Yiqi Huyu Qingre prescription and its separates,Tripterygiumwilfordi can affect the expression of NF-κB and IκB-αof the MsC cultured invitro.About the results of restraining the expression of NF-κBp65, the effect ofhuayu serum group was the strongest,when enhanced the expression ofIκB-α,qingre serum group was stronger than huayu serum group and yiqi serumgroup,and yiqi serum group was the weakest.Different groups of Chinese medicinemay choose different target function, strength is also different, regarding themechanisms still need further research;in the comparison of the compound Chinesehuayu and qingre group were obviously better than the effect of yiqi group,therefore, in the treatment of proliferative glomerulonephritis should emphasis onpromoting blood circulation to remove blood stasis and clearing away heat andtoxic material.
