Objective:Objective to detect the expression of GRIM-19and STAT3inosteosarcoma by S-P immunohistochemical method,and their relationshipwith clinical pathological characteristics of the patients and relativity betweenthe expression of the two factors in osteosarcoma,and provide certainreferences for clinical diagnosis and treatment of osteosarcoma.Methods:1. All the specimens in vitro within two hours were placed in10%neutralformalin and fixed for24-48hours,fixed samples were made of wax after theprocess of dehydration,vifrificationt,putting into the wax and embedding,4umserial section,take1for routine HE staining to confirm pathological diagnosisagain,the others were used to STAT3and GRIM-19streptomycinavidin-peroxidase method (Streptavidin peroxides conjugated method,SP)staining.Known positive plate as a positive control, PBS instead of the firstantibody as negative control.2. The expression of GRIM-19and STAT3were detected in35cases ofosteosarcoma and14cases of osteochondroma by immunohistochemical S-Pmethod,and the operation was in accordance with the kit instrution,GRIM-19、STAT3polyclonal antibody working concentration were respectively1:50,1:100.3. Statistical analysis: all the data were analyzed by SPSS17statistic,Fisher exact probability test and the chi square test were used to count datasample rate,Spearman rank correlation analysis was used to count therelationship of the two factors.P <0.05was statistically significant difference. Results:1. Expression of STAT3and GRIM-19in osteosarcoma, osteochondroma:the positive expression rate of STAT3in osteosarcoma was significantlyhigher than that of osteochondroma, the positive expression rate of GRIM-19was significantly lower than that of osteochondroma, the differences werestatistically significant (P <0.05).2. The relationship between STAT3,GRIM-19expression andclinicopathologic features of osteosarcoma: the expression of STAT3inOsteosarcoma has nothing to do with age, sex (P>0.05), but associate withEnneking staging, lung metastasis, the positive expression rate of STAT3increased gradually from Enneking Ⅰ to Ⅲ period,positive expression ofSTAT3was significantly higher in Osteosarcoma of lung metastasis than thatof no pulmonary metastasis (P <0.05). The expression of GRIM-19inOsteosarcoma has nothing to do with age、sex、pulmonary metastasis (P>0.05), but associate with Enneking staging, the positive expression rate ofSTAT3decreased gradually from Enneking Ⅰ to Ⅲ period (P <0.05).Expression of GRIM-19relate to Enneking stage of osteosarcoma.3. The relationship between the expression of GRIM-19and STAT3inosteosarcoma:the expression of STAT3is negatively related to and that ofGRIM-19in osteosarcoma (r=-0.528, P <0.05).Conclusion:1. The positive expression of STAT3in osteosarcoma was significantlyhigher than that of osteochondroma,and it related to whether patients withosteosarcoma pulmonary metastasis and Enneking stage,it illustrate that theexpression of STAT3is bound up with clinic progression and invasion ofosteosarcoma.2. The positive expression rate of GRIM-19in osteosarcoma wassignificantly lower than that of osteochondroma,and it related to Ennekingstage of patients with osteosarcoma,it showed that the expression of GRIM19was related to the clinic progression of osteosarcoma,and it is an importantevent in the occurrence of osteosarcoma.3. The expression of STAT3was negatively correlated with GRIM-19in osteosarcoma,it showed that high expression of STAT3and low expression ofGRIM-19coexist in osteosarcoma tissues,two common function may lead totumor cells more susceptible to deterioration and abnormal proliferation,thuscontributing to the occurrence and development of osteosarcoma.
