| Objectives:To evaluate the feasibility and treatment of spinal cord injury by transfected and recombinant adenovirus containing GDNF gene of rat bone marrow mesenchymal stem cells with chitosan.Methods:(1) the preparation of recombinant adenovirus carrying GDNF gene which from rats.(2) Group A:only bMSCs. Group B:Adenovirus transfection group without GDNF.Group C: Adenovirus transfection group with GDNF.Group D:Adenovirus transfection group with GDNF and chitosan.(3) Injection the material of each group into the position of spinal cord injury in SD rats, and related detection recovery motion system of rats were observed after operation. Results:(1) Group A:bMSCs growing normally. Group B:BMSCs expressing green fluorescence but no change in morphology Group C:BMSCs expressing green fluorescence and changing in morphology.Group D:The result is the same as group C, but expressed for a long time.Each group was treated SCI rats:A rats compared with no intervention blank rats in group SCI BBB score improved, but not ideal. B group of related detection with no significant differences between group A. Compared with control group and A C group, B group showed significant improvement. The rats in group D were better than those in group C reply.Conclusion:. Duration of enhancement green fluorescent recombinant adenovirus was transfected into bMSCs with transfection efficiency and longer larger carrying GNDF gene, sustainable GDNF release neurotrophic factor, induce the differentiation of bMSCs, at the same time, composite chitosan nano scaffold transplantation for treatment of spinal cord injury has certain feasibility. |
