Objective:Through observing the different effect of Rhizoma Ligusticum and Gastrodia elata with different dosage of compatibility in migraine rat model with hyperactivity of liver-yang to explore the rules of compatibility of Rhizoma Ligusticum and Gastrodia elata. Further, to give Chinese medicine clinical suggestions about using drug pairs with rise and fall, check and balance.Totally80SD rats were randomly divided into8groups with10rats:blank group (Z), model group (M), Rhizoma Ligusticum and Gastrodia elata with (4:1、1:1、1:4) pro-portions group extracted by water(A、B、C), Rhizoma Ligusticum and Gastrodia elata with (4:1、1:1、1:4) pro-portions group extracted by alcohol (D、E、F). Except model group(Z), all the rest are irrigated Aconite decoction which contain0.2g/ml drug lasting21d with2g cto duplicate hyperactivity of liver-yang model. The blank group are irrigated distilled water with same bulk lasting21d at the same time.Rats in A、B、C、D、E、F groups are irrigated Rhizoma Ligusticum and Gastrodia elata with (4:1、1:1、1:4) mathing up the same pro-portion and extracting medicinal liquid which contain0.175g/ml drug lasting7d with1.75g μg·kg-1·h-1beginning at the15th d of irrigating Aconite decoction, to give preventive drug before duplicating migraine model. The blank group are irrigated distilled water with same bulk lasting7d at the same time. At the seventh day, except blank group all the rest are injected subcutaneously nitroglycerin according10mg/kg at the right shoulder after irrigating drug. The blank group are injected subcutaneously physiological saline solution at same the right shoulder, bulk lasting7d at the same time. After sc nitroglycerin30min, every group give drug one time excluding Z and M.All rats are anaesthetized after sc nitroglycerin4h. Then draw blood from aorta of abdomen and centrifuge blood at3500r/min for15min at4℃. Separate serum from centrifugal blood and keep it in-20℃refrigerator to determine CGRP、NOS、NO、ET、TXB2、6-Keto-PGF1a and the ratio of NO and E1, TXB2and6-Keto-PGF1a in serum.. Concoct reagent and operate procedure went strictly according to the instructions. Use SPSS17.0analyse experimental data with variance. P<0.05is regarded as an statistical meaning.Result:(1) The effect of Rhizoma Ligusticum and Gastrodia elata with different dosage of compatibility in migraine rat model with hyperactivity of liver-yang about CGRP in rat’s serum.Compare among groups extracted by alcohol, Rhizoma Ligusticum and Gastrodia elata with pro-portions (1:4) group has the best effect in these groups about reducing CGRP content (P<0.05-0.01). The (4:1) group has better effect than (1:1) group (P<0.05). These result declare that Rhizoma Ligusticum and Gastrodia elata with different dosage of compatibility have significant different effect about CGRP content in migraine rat model with hyperactivity of liver-yang. And the effect from good to bad in sequence is (1:4)>(4:1)>(1:1)(2)The effect of Rhizoma Ligusticum and Gastrodia elata with different dosage of compatibility in migraine rat model with hyperactivity of liver-yang about NOS、NO、ET NO/ET in rat’s serum.Compare among groups extracted by water, Rhizoma Ligusticum and Gastrodia elata with pro-portions (1:4) group has the best effect in these groups about reducing NO content and NO/ET (P<0.05-0.01). The (1:1) group has better effect than (4:1) group about reducing NO/ET (P<0.01). The (1:1) group has better effect than (4:1) group about rising ET content in serum.(P<0.05).Compare among groups extracted by alcohol, Rhizoma Ligusticum and Gastrodia elata with pro-portions (1:4) group has the best effect in these groups about reducing NOS activity (P<0.05-0.01). The (1:1) group has better effect than (4:1) group about reducing NOS activity (P<0.01).(1:4) group has better effect than (1:1) group about reducing NO content (P<0.05).In short, comparing the effect of Rhizoma Ligusticum and Gastrodia elata with different dosage of compatibility in migraine rat model, the (1:4) group has the best effect about NOS、NO、ET、NO/ET in serum regardless of extracted methods.(3)The effect of Rhizoma Ligusticum and Gastrodia elata with different dosage of compatibility in migraine rat model with hyperactivity of liver-yang about TXB2、6-Keto-PGF1a、TXB2/6-Keto-PGF1a、in rat’s serum.Compare among groups extracted by water, Rhizoma Ligusticum and Gastrodia elata with pro-portions (1:4) group has the best effect in these groups about rising6-Keto-PGF1a content (P<0.05-0.01). The (1:1) group has better effect than (4:1) group about rising6-Keto-PGF,a content (P<0.01). The (1:4) group has the best effect in these groups about reducing TXB2/6-Keto-PGF1a (P<0.05-0.01).Compare among groups extracted by alcohol, Rhizoma Ligusticum and Gastrodia elata with pro-portions (1:4) group has the best effect in these groups about rising6-Keto-PGF1a content (P<0.05-0.01). The (1:1) group has better effect than (4:1) group about rising6-Keto-PGF1a content (P<0.05). The (1:4) group has better effect than (1:1) group about TXB2/6-Keto-PGF1a (P <0.05).In short, comparing the effect of Rhizoma Ligusticum and Gastrodia elata with different dosage of compatibility in migraine rat model, the (1:4) group has the best effect about6-Keto-PGF1a、TXB2/6-KetoHPGF1a in serum regardless of extracted methods.(4) The different effect of Rhizoma Ligusticum and Gastrodia elata in same proportion with different extracted methods in migraine rat model with hyperactivity of liver-yangSum up the result of comparing the same proportion extrated by different method, the effect of Rhizoma Ligusticum and Gastrodia elata with different pro-portions groups extracted by alcohol has better effect than by water in migraine rat model with hyperactivity of liver-yang(P<0.05-0.01).Conclusion:After comparing all factor, it can be confirm that there are significantly different effects of Rhizoma Ligusticum and Gastrodia elata with different dosage of compatibility in migraine rat model with hyperactivity of liver-yang. |
