| Objective:This study aimed to study the role of Lipoic acid in treat the high OS in ratswith diabetic nephropathy(DN),to research the relationship between AGEs and OS. This studyalso investigate the function of Lipoic acid on RAGE, blood glucose, insulin resistance,andblood lipids.Methods: Randomly choose6rats from30healthy female wistar rats to be normalcontrol group(group A).The rest24rats treated by high-sugar-fat feed for8weeks, managewith streptozocin by twice intraperitoneal injection, the successful model of the rats wererandomly divide into diabetic nephropathy group(group B) and lipoic acid group(group C)according to the blood sugar levels. Give group B saline(50mg.kg-1.d-1) by intraperitonealinjection for4weeks while group C treated by lipoic acid(50mg.kg-1.d-1).Assay and analysisdatas of all subjects, such as random blood sugar, FPG,24hours urinary albumin, TG, TC,HDL-C, LDL-C, MDA, GSH-Px, SOD, glycosylated hemoglobin(GHbA1c), AGEs,andkidney RAGE gene expression.Results: Aggregately8cases were excluded because of failing model or death, and22entered the final statistical analysis.1.B and C group’s HbA1cã€FPGã€FINSã€AGEs significantlyhigher than those in group A(P<0.05);Group C have low level HbA1cã€FPGã€FINSã€AGEs(P<0.05) compare with group B.2. Compared with group A,B and C group’s TCã€TGã€LDL-Cã€24hours urinary albumin are significantly higher(P<0.05); Group C have low levelTCã€TGã€LDL-Cã€24hours urinary albumin (P<0.05) compare with group B. There was nodifference in HDL-C.3. Compare with group A, the MDA of group B increases(P<0.05),whileSODã€GSH-Px decreased; Compare with group B, the MDA of group C decreased whileSODã€GSH-Px increased(P<0.05).4.Diabetic nephropathy group and LA group were detectedthe expression of RAGE in renal tissue, and the consequence of normal control group isnegative.Conclusions:1. Diabetic nephropathy rats have higher AGEs and MDA while SODdecreased, and it increase the expression of RAGE in renal tissue, indicating the interactionbetween AGEs and RAGE in cell signal transduction pathway perhaps play an important role in the process of renal injuries.2.Lipoic acid can inhibit oxidative stress, reduce AGEs andRAGE expression level in kidney tissue. It also reduce urinary protein. |
