Objective:Hesperidin was extracted from citrus flavonoids, which has strong biological activity.Our research group carry out a series of studies about hesperidin.Tea polyphenol is the typical natural product extracts recently, has been highlighted in the field of oral.The aims of this study were to investigate the effects of hesperidin on tooth hard tissue demineralization and remineralization, and compare it with tea polyphenols and chlorhexidine.To provide theoretical and experimental basis for hesperidin and tea polyphenol’s clinical application.Methods:1.Sample preparation of artificial caries:Prepared enamel and dentin samples were placed into demineralization solution for96hours at37℃.Then formed artificial caries specimens.2.The experiment group:Experimental groups:5000ppm HPN;5000ppmHPN+1000ppmNaF;2000ppmTP;2000ppmTP+1000ppmNaF;2000ppmCHX.Positive control:deionized water.Blank control:1000ppmNaF.3.pH cycling experiment:Each group of samples were immersed in demineralization solution14h,test solution2h and remineralization solution8h at37℃.Each group of samples were thoroughly rinsed between each immersion with buffer solution5min before treatment.The pH cycling was performed for8days.4.Observation indexes:Use the release value of calcium,phosphorus ions as the index to evaluate the drug to the specimen demineralization of influence.Use the surface microhardness recovery value and the surface of a sample of calcium and phosphorus atom percentage and ratio of calcium and phosphorus,and hydroxyproline concentration as the index to evaluate the effect of re mineralization.Results:1.The release value of calcium, phosphorus ions in demineralization solutionEnamel:There wasn’t statistically significant when HPN,TP and CHX compared with the blank control group (p>0.05)Dentin:There wasn’t statistically significant when HPN,TP and CHX groups of calcium release value compared with the blank control group (p>0.05).HPN group of phosphorus release value were lower than the blank control group (p<0.05)2. Ca/PHPN,TP and CHX groups of enamel and dentin specimen were higher than the blank control group,the difference was statistically significant (p<0.05)3.Surface microhardnessHPN,TP and CHX groups of enamel and dentin specimen compared with the blank control group, the difference was not statistically significant (p>0.05).4.Hydroxyproline concentration in remineralization solutionHPN and CHX group were lower than the blank control group (p<0.05); TP group compared with blank control group there wasn’t statistically significant (p>0.05)5.Combined with NaFHPN+NaF and TP+NaF group compared with blank control group there was differences statistically significant (p<0.05)Conclusions:1.HPN, TP and CHX can promote remineralization in enamel.2.HPN has the function to inhibit dentin demineralization; has the inhibitory effect of collagen matrix degradation; has the function to promote dentin remineralization.3.TP has the function to stable crystal structure and promote dentin remineralization.4.CHX has the function to inhibit collagen matrix degradation and promote dentin remineralization.5.HPN and TP combined with NaF have no synergistic effect.6.Compared with the surface microhardness testing,spectral analysis is a more sensitive indicator of the remineralization.
