Objectives:To investigate the expression of CYP1B1and COX-2in invasive breast cancer and in ductal carcinoma in situ (DCIS) and benign lesions and ca-adjoining normal breast epithelium, and assess the relationship between the expression of CYP1B1and COX-2and clinieopathologie characteristic, laying a pathologic foundation for breast cancer understanding of their roles in the tumorigenesis of breast cancer cells.This study will also observe the expressions of Vimentin and Bcl-2in molecular subtypes of breast cancer and their correlation.in ordor to provide a new basis for classification and screening of high invasive breast cancer patients.Methods:1. Expression of CYP1B1and COX-2in Human Breast Cancer and Its Significances110paraffin-embedded samples of breast tissue were collected and divided them into4groups:invasive breast cancer, ductal carcinoma in situ (DCIS), Benign lesions and Ca-adjoining normal breast epithelium. Immunohistochemieal method was applied to check the expression of COX-2and CYP1B1protein in110cases of breast tissue.Analyze the relationship between CYP1B1and COX-2and clinicopathological parameters.2. The Expression and correlation of Vimentin and Bcl-2in molecular subtypes of Breast CancerImmunohitochemical study for ER, PR and HER-2was performed on56cases of breast cancer tissue, and divided them into4molecular genetic subgroups:luminal A (ER+and/orPR+, HER-2-), luminal B (ER+and/orPR+,HER-2+),HER2/neu-positive (ER-and PR-, HER-2+), and triple-negative (TNBC)(ER-and PR-, HER-2-). The immunohitochemical method was used to detect the expression of Vimentin and Bcl-2.The relationship between the clinicopathological characteristics and the expression of Vimentin and Bcl-2were analyzed, so was the correlation between the expression of Vimentin and Bcl-2. Results:1. Expression of CYP1B1and COX-2in Human Breast CancerIn invasive breast cancer, DCIS, benign lesions and ca-adjoining normal breast epithelium, CYP1B1and COX-2expression rates were (77.8%,69.2%,90.9%) and (74.0%,92.3%,87.5%and14.3%) respectively, no significant difference in the positive expression rate of CYP1B1and COX-2was found between invasive cancer, DCIS, benign lesions group, However, compared with the above three groups, the positive expression rate of CYP1B1and COX-2was significantly decreased in cancer-adjoining normal breast epithelium (P<0.001). There was a significant correlation between expression of COX-2and CYP1B1in breast cancer(r=0.250, P=0.046).Comparison between CYP1B1and COX-2in breast cancer showed that there was no statistically significant difference with axillary lymph node metastasis and histologic grade(P>0.05), whereras expression of CYP1B1correlated with ER and PR(P<0.05).2. The Expression and correlation of Vimentin and Bcl-2in molecular subtypes of Breast CancerIn luminal A, luminal B, HER2/neu-positive, and TNBC, Vimentin and Bcl-2expression rates were (29.1%,33.3%,66.7%,81.3%) and (79.2%,77.8%,33.3%,37.5%) respectively,, no significant difference in the positive expression rate of Vimentin and Bcl-2was found between luminalA and luminalB subgroup, so as HER2/neu-positive and TNBC. However, compared with the above four groups, the positive expression rate of Vimentin was significantly increased in HER2/neu-positive and TNBC (P<0.01) while the positive expression rate of Bcl-2was significantly decreased in HER2/neu-positive or TNBC(P<0.05). In breast cancer subgroups, the positive expression rate of Vimentin and Bc1-2were significantly negatively correlated(r=-0.287, P<0.05).Conclusions:1. Compared with cancer-adjoining normal breast epithelium, positive expression rate of CYP1B1and COX-2in invasive cancer, DCIS and benign lesions groups increased significantly. Both COX-2and CYPIB1express and function in human breast tumorigenesis, COX-2may play a positive role in upregulating CYPIB1.2. Given that there was no statistically significant relation between CYP1B1/COX-2expression and aggressive phenotypic features, such as high histological grade, large tumour size, lymph node positivity, age and ER/PR status, which are considered prognostic parameters, suggests that CYP1B1and COX-2expression cannot have a place among the other prognostic parameters of breast carcinoma.3. In molecular subtypes of breast cancer,Vimentin expression was abnormally increased and the Bcl-2expression was abnormally decreased in HER2/neu-positive and TNBC. Both probably showed a correlation with poor prognosis.The detection of Vimentin and bcl-2is of great importance in grading subgroup of breast cancer and evaluating the prognosis and screening the patients of high aggressive risk.
