| Objective:To explore the relationship between ASIC3expression and the rat bladder detrusor activities. Determining the relationship between the protein of ASIC3and urinary irritation. Provide a new research for urinary irritation and overactive bladder.Methods:Select adult female rats, use cyclophosphamide (CYP) intraperitoneal injection or e. coli bladder perfusion to induce OAB rats model and bacterial cystitis model.According to the different method of building,all the rats are divided into:1, CYP team,2, bacteria team.Then divided into the flowing groups:A1:normal group, B1:OAB group, C1:OAB control group (OAB+amiloride);A2:normal group, B2:cystitis gtoup, C2: cystitis control group (cystitis+amiloride). The six groups of rats bladder are pour into saline or amiloride saline solution, then do the urine flow dynamic test.Take six groups of rats bladder tissue at the same time for the pathology examination.At last,through immunohistochemical staining, rt-pcr and western-blot method to measure the content of ASIC3differencesResults:Urine flow dynamic tests show:normal group have no spontaneous activeties during the urine storage period and urination period; OAB groupã€cystitis group and the two control groups have spontaneous activeties during the urine storage period and urination period. Compared with the normal group:the urination interval are significantly shortened (P<0.01), micturition number increased significantly (P<0.01); Compared with OAB group, the spontancous activetie in OAB control group is less,the micturition number decrease and the urination interval is longer(P<0.05); Compared with cystitis control group,the spontancous activetie in cystitis control group is less,the micturition number decrease and the urination interval is longer(P<0.05);Pathology detections show that OAB group, cystitis, OAB intervention group and cystitis intervention group rat bladder mucosas are injured; Immunohistochemical shows:rats bladder tissue containing ASIC3; And ASIC3mainly exist in the bladder mucosa, ASIC inhibitors can inhibit bladder mucosa ASIC3expressing; muscle layer a small amount of ASIC3.ASIC3inhibitors have no obvious effect on muscle layer.Rt-PCR and western-blot show:the expressing of ASIC3in bladder in OAB group and cystitis group increased significantly (P<0.01), after adding ASIC3inhibitors, the expressing of ASIC3in bladder in OAB group and cystitis group decreased(higher than normal group,P<0.05; lower than OAB group and cystitis group,P<0.05).Conclusion:1, the rat bladder contains ASIC3and ASIC3mainly exist in the bladder mucosa;2, The ASIC3in OAB rats model and cystitis rats bladder are higher than normal rats, the expression is influenced by ASIC inhibitor obviously.3, Thw rising of ASIC3expressing in bladder may be closely related to urinary irritation symptoms or OAB. |
