Aging Osteoporosis:Correlation With Tregs And Inflammatory Factors

Objective We want to explore the correlation of osteoporosis with low-grade inflammation and Tregs.the immunomodulatory mechanism of age-induced osteoporosis by measuring the absolute value of Tregs in peripheral blood and the concentration of serum RANKL, OPG OPG, TNF-a, IL-1, IL-6and bone mineral density of different age groups of people.Methods The244health subjects(between35to75years old)were divided into four groups for10years old as a group. BMD of the post anterior lumbar spine(L1、L2、L3、L4、L1-4) were examined by dual-energy xray absorptiometry (DEXA), and the proportion of Tregs was detected by flow cytometry. The concentration of RANKL、OPG、TNF-α、IL-1and IL-6in serum were measured by enzyme-linked immuno sorbent assay(ELISA).Results1.The BMD of Lumbar in four age groups(35-44.45-54、55-64、65-75years old) are different, and is decreasing with increasing age (P=0.000)2. peripheral blood Ca2+, P3-, PTH, VD, TC, TG, LDL-C in different age groups are not significant different (P>0.05);Osteocalcin in different groups is different,and it is decreasing with increasing age(P=0.039).DPD in different groups is different, and it is increasing with increasing age(P=0.000). BALP in different groups is different,and it is decreasing with increasing age(P=0.000).3. Tregs,RANKL,OPG,RANKL/OPG,IL-1and IL-6in different groups are different (P<0.05). Tregs,RANKL,OPG are decreasing with increasing age and IL-1,IL-6,RANKL/OPG are increasing with increasing age (P<0.05).TNF-a in different groups is not significant different (P>0.05).4. Tregs, RANKL, OPG are positively correlated with BMD of L1, L3, L4,L1-4(P<0.05),and they are not significant correlated with BMD of L2(P>0.05). RANKL/OPG,IL-1are not significant correlated with BMD of L1,L2,L3,L4,L1-4(P> 0.05).TNF-a is negatively correlated with BMD of L2,but it is not significant correlated with BMD of L1,L3, L4, L1-4(P>0.05).IL-6is negatively correlated with BMD of L1, L2,L3, L4, L1-4(P<0.05)5. Tregs is not significant correlated with RANKL(P>0.05).Conclusions1. Bone resorption is increasing with increasing age, bone formation and bone density is decreasing with increasing age.2. Reducing the number of Tregs may contribute to the osteoporosis with increasing age. Whether this role of Tregs is related with RANKL/RANK/OPG system still needs further study.3. IL-6, TNF-α may stimulate the bone resorption by increaseing the formation and differentiation of osteoclast. Whether age can promote osteoporosis by affecting inflammatory factors remains to be further studied.4.RANKL/RANK/OPG system enhance bone resorption with increasing age, thereby promoting osteoporosis.
