BackgroundProstate cancer (PCa) is one of the common disease in male genitourinary system. At present, the treatment of prostate is the main operation and endocrine therapy, because prostate cancer patients are elderly patients,and early symptoms are not obvious, therefore, the majority of patients the presence of cancer surrounding tissue invasion and metastasis, so lose the chance of radical operation. Endocrine therapy can alleviate the condition, but easy to relapse.Therefore, the search for a new method of treatment has become a key factor in the survival of prostate cancer patients,at the same time, may also provides a new reference to the treatment and prognosis of prostate cancer.ObjectiveIn this study, patients with adenocarcinoma of the prostate and normal hyperplasia prostate tissue as the research object, To investigate the expression level of hypoxia-inducible factor-1and glucose transporter-1by immunohistochemical method, and analyze its relationship with the prostate cancer clinicopathologic significances; at the same time, Through this research we hoped to explore the correlation between the HIF-land GLUT-1in prostate adenocarcinoma, and discuss their role and clinical application value.MethodsCollect the specimen from February2012to August2013in First Affiliated Hospital of Xinxiang Medical College Department of Urology. Among the46cases of adenocarcinoma of the prostate specimens, including12cases of retropubic prostatectomy;34underwent transurethral resection of the prostate. Specimens were confirmed by pathological diagnosis of prostate cancer, preoperative chemotherapy and radiotherapy were not; Another40cases of benign prostatic hyperplasia group as controls. The organization uses fixed in paraformaldehyde, dehydrated, embedded in paraffin,3um serial sections, and drying, dewaxing, hydration, immunohistochemical staining (DAB), detecting the expression level of HIF-1a and GLUT-1of prostate cancer and prostatic hyperplasia tissue,and to investigate their relationship and correlation with the pathological grade and clinical stage of prostate cancer. Each slice was randomly selected non-overlapping five high-power fields, respectively, by two observers who do not know the slice grouping, observe and count the number of positively stained cells. Value method using the AB (semi-quantitative method) for analysis.ResultsImmunohistochemical DAB method showed that positive expression rate of HIF-la and GLUT-1protein in prostate adenocarcinoma was60.9%and67.4%respectively; prostatic hyperplasia its positive expression rate was12.5%and17.5%respectively. Comparing this two, HIF-la and GLUT-1expression in prostate adenocarcinoma were significantly higher, the difference was statistically significant (P<0.05);HIF-la and GULT-1expression in prostate cancer patients with clinical and pathological grading and staging were as follows:in poorly differentiated tumor grade, infiltration and lymph node and distant metastasis specimens HIF-la and GLUT-1the positive rate of expression were significantly higher than well-differentiated, non-metastatic expression of both specimens (P<0.05). The expression of HIF-la and GLUT-1were a significant positive correlation in prostate adenocarcinoma tissues(P<0.05). Conclusions1. High expression of HIF-1a and GLUT-1are positively correlated in prostatic adenocarcinoma tissue, and closely related with clinical pathological characteristics of the tumor, it may play an important role in the occurrence and development of prostate cancer.2. HIF-la and GLUT-1may be considered the biological behavior index of prostate adenocarcinoma.
