Objective:To investigate the changes of epinephrine (EPI) and tumor necrosis factor alpha (TNF-a) in serum in pulmonary injury induced by mechanical trauma in non-lethal mechanical trauma rat model and their relationgship with pulmonary injury and to inquire into the possible mechanism and clinical significance.Methods:Forty Wistar rats were randomly divided into the pseudo-trauma group (P), trauma group(M), normal control group (C), epinephrine hydrochlaride group (E) and tumor necrosis factor alpha group (T), there are eight in every group. The first part, the rat model of non-lethal mechanical trauma group was caused by the Noble-Collip trauma instrument, which includes pseudo-trauma group and trauma group; the second part, and the pharmaca pulmonary injury model group was caused by the following methods, epinephrine hydrochloride and tumor necrosis factor-a were injected into the rats intraperitoneally with a certain volume, and an equal volume of normal saline were injected into rats intra-peritoneally as normal control group. All rats were killed in the successful modeling of6h by abdominal aortic blood, HE staining observes the lung tissue pathology, terminal deoxynucleotidyl transeferase-mediated nick end-labeling (TUNEL) detected the apo-ptosis index in lung tissue, the serum levels of change of epinephrine and TNF-a were asssyed by ELISA.Results:1The changes of epinephrine and TNF-a in serumThe test results of Adrenaline content in serum:Trauma group (1.7306±0.1394) ng/ml compared with the pseudo-trauma (0.1712±0.0193) ng/ml were significantly higher, the difference was statistically significant (p<0.01). The test results of TNF-a content in serum:Trauma group (127.2135±1.5018) ng/L compared with the pseudo-trauma (10.9745±1.0042) ng/L were also significantly higher, the difference was statistically significant (p<0.01)2Histopathology changeThe pseudo-trauma group and control group were gene-rally observed no abnormal above the rat’s lung surface, Trauma group, epinephrine Hydrochlaride group and tumor necrosis factor alpha group have a little or a sheet bleeding of lung surface with varying degrees of swelling and congestion.The observation of ordinary light microscope, the alveolar structure of the pseudo-trauma group and normal control group are normal and continuous, the interstitial and alveolar have no obvious inflammatory cells and red blood cells, trauma group, epinephrine hydrochlaride group and tumor necrosis factor alpha group were significantly bleeding and inflammation in the lung tissue of mechanical trauma rats, the alveolar wall structure were subjected to different degrees of damage, there are have some exudation, hyperemia, hemorrhage and varying amounts of a variety of inflammatory cells infiltration in the interstitial and alveolar of mechanical trauma rats.3The analytic results of TUNELThe trauma group, epinephrine hydrochloride group, and tumor necrosis factor-a group showed obvious apoptosis of lung cells, but the pseudo-trauma group and control group have no significant apoptosis of lung cells.Conclution:The increased content of epinephrine and TNF-a in serum significantly increased the lung tissue inflammation and apoptosis index in lung tissue, which indicated that may be concerned with pulmonary injury caused by non-lethal mechanical trauma.
