Objective Comparing the expression of serum proteins in women with preeclampsia and normal pregnancy with Isotopically labeled relative and absolute quantification (iTRAQ) technology and Combined liquid chromatography tandem mass spectrometry (LC-MS/MS) technology to screen out the potential plasma biomarkers,and to explore the possible mechanism of them in the development of pre-eclampsia.Methods From Jun2012to Jun2013the women with severe pre-eclampsia (SPE group) and mild pre-eclampsia (MPE group) were collected from the clinic among the pregnancy women who diagnosed and regular checked in Affiliated Hospital of Qingdao University for this study.Each group has30cases.Additionally the normal pregnant women (control group)were collected as control group.Blood was collected and proteins were extracted from serum.Then,albuminous degeneration,reduction and enzymolysis were performed in SPE group and control group. iTRAQ was adopted to achieve the expression difference. Then we Selected two of the differentially proteins Gelsolin,S100A8with ELISA and real-time quantitative PCR for further verification.Results①The difference proteins in plasma can effectivity identified by iTRAQ combined with LC-MS/MS technology were234,of which24proteins were identified in SPE group more1.5times(increase ratio>1.50or lower ratio<0.67) than the control group by T-test,of which14points up regulation of preeclampsia, and the other ten points down regulated.②The plasmas levels of Gelsolin in SPE group(0.363±0.152) and MPE group (0.288±0.013) significantly higher than that in control group(0.204±0.011).③The plasmas levels of S100A8in SPE group (0.928±0.101) and MPE group (0.687±0.064) significantly higher than that in control group(0.550±0.073).④The expression of Gelsolin mRNA in placenta in SPE group (4.42±1.059) and MPE group(2.44±0.614) had significantly difference with control group(0.90±0.037),(P<0.05).⑤The expression of S100A8mRNA in placenta in SPE group (7.59±1803) and MPE group(3.83±0.879) had significantly difference with control group(0.98±0.04),(P<0.05).⑥In preeclampsia,plasma Gelsolin level had no correlation with placental Gelsolin (r=0.163, P>0.05);⑦In preeclampsia,Plasma S100A8level had significant association placenta S100A8(r=0.701, P<0.05).Conclusions iTRAQ combined LC-MS/MS technology can effectively filter out the difference proteins between preeclampsia and normal pregnancy. There are some relationships between Gelsolin、S100A8and the development of preeclampsia.S100A8may be used as preeclampsia specific early serum protein markers,lay a theoretical foundation for the early diagnosis of preeclampsia.
