| Objective To investigate the expression of soluble vascular adhesionprotein-1(sVAP-1) on peripheral blood and its clinical significance in idiopathic pulmonaryfibrosis(IPF).Method sVAP-1, TGF-β1, TNF-α and IL-6expression on peripheral blood from26IPF patients,18non-idiopathic pulmonary fibrosis-interstitial lung disease patients(non-IPF-ILD) and30healthy controls (HC) was examined by using enzyme-linkedimmunosorbent assay (ELISA). The degree of pulmonary fibrosis in patients with IPFwere determined by high resolution CT(HRCT) and the lung function such as forced vitalcapacity (FVC), total lung capacity (TLC) and diffusing capacity of CO (DLCO) wasrecorded. Data were analyzed with t test and Pearson correlation test.Results The level of expression of sVAP-1in IPF patients (732.5±361.8ng)peripheral blood was significantly increased compared to non-IPF-ILD patients (471.4±207.5ng,p<0.01,t=2.76) and normal controls(409.9±148.3ng,p<0.001,t=4.48). But therewas no significant difference of sVAP-1between non-IPF-ILD patients and normalcontrols. The concentrations of TGF-β1〠TNF-α and IL-6(respectively,729.1±243.9pg/ml,8.9±3.3pg/ml,78.9±19.3pg/ml) were significantly higher in IPFpatients than in normal controls(respectively,316.5±107.2pg/ml,6.7±2.5pg/ml,58.7±20.9pg/ml)(all p<0.05).18IPF patients in several pulmonary fibrosis group (1033.0±424.8ng/ml) displayed a higher expression of sVAP-1than mild pulmonary fibrosisgroup (598.8±238.0ng/ml)(p<0.01,t=3.36). The level of expression of sVAP-1onperipheral blood from IPF patients was inversely correlated with FVC/Pred and DLCO/Pred(both p<0.05). The level of expression of sVAP-1on peripheral blood from IPFpatients was positively correlated with the concentrations of TGF-β1, TNF-α and IL-6(allp<0.05).Conclusion The expression of sVAP-1on peripheral blood from IPF patients isincreased and inversely correlated with lung function and is also positively correlated withthe concentrations of TGF-β1, TNF-α and IL-6. Increased expression of sVAP-1mightplay an important role in the pathogenesis of IPF. |
