Objective:To discuss the inflammation and oxidative stress indicators on atherosclerosis in rats, then research the effect of Rosuvastatin and Probucol on these indicators in serum.Methods:Fifty-four male Wistar rats were randomly devided into5groups, Normal diet control group (Group A) has12rats, High-fat diet group (Group B) has12rats, Rosuvastatin group (Group C) has10rats, Probucol group(Group D) has10rats, and Rosuvastatin+Probucol group(Group E) has10rats. Normal diet control group was given normal diet, high-fat diet group and drug intervention group were given high fat diet and egg albumin(2.5mg/kg) by intraperitoneal injection (5times/week). Drug intervention was given to GroupC,D,E from the beginning of the ninth week. At the end of the sixteenth week, all the rats were weighted and anethetized, and blood samples were collected from right atrium, then detect the indicatots of VE-cadherin,8-iso-PGF2a,ANP and SOD. The histomorphological changes of the aorta were observed under light microscope.Results:Compared with Group A, the body weight in GroupB,C,D,E decreased significantly (P<0.01), VE-cadherin and8-iso-PGF2a in serum were significantly higher (P<0.01), ANP and SOD were decreased(P<0.01); Compared with Group B, the body weight in GroupC,D,E were heavier(P<0.05), the levels of VE-cadherin and8-iso-PGF2a in serum were significantly lower (P<0.01), APN and SOD were elevated(P<0.05), the endothelial damage of the aorta were less severe; Compared with Group C, the level of8-iso-PGF2a in serum was lower in Group D,E(P<0.01), and the level of SOD was elevated (P<0.01); Compared with Group C, the level of VE-cadherin in serum was lower in Group E (P<0.01), and the level of APN was elevated(P<0.01); Compared with Group D, the level of VE-cadherin in serum was lower in Group E (P<0.05), and the level of APN was elevated(P<0.05); Compared with Group C,D, the intimal thickness in Group E was thinner(P<0.05).Conclusion:Resuvastatin and Probucol have the effect of anti-inflammation and antioxidation, but the antioxidation of Probucol was superior to Rosuvastatin. Combination of both can decrease the level of VE-cadherin and8-iso-PGF2a in serum,and increase the activity of APN and SOD, and decrease the histomorphological damage.The results of this experiment can guide the rational use of both Rosuvastatin and Probucol in clinical practice.
