Objective To research the gene expression profiles of ischemic postconditioning on renal ischemic reperfusion injury.Methods60SD rats were allocated randomly to6groups:control group(s, n=10), Ischemia-reperfusion(IR) group and ischemic postconditioning group of A, B, C, D.Choose the better model of the ischemic postconditioning by the blood samples and nephridial tissue section.Then,6SD rats were allocated randomly to2groups:Ischemia-reperfusion group and ischemic postconditioning (IR-P) group.Group IR:gature the right renal pedicle. Left renal pedicle was clamped for60min and then reperfused. IR-P group:Left renal pedicle was clamped for60min and reperfused after6cycles of10s ischemia and10s reperfusion.Raised after24h quickly cut the left kidney and put the left kidney tissue in liquid nitrogen cryopreservation, respectively.By RNA extraction,probe preparation, chip hybridization, sweep signal and collect image,use Softfire Decisionsite8.0software to analyze the gene expression.According to the function of the gene encoding protein analyze genes differentially expressed more than twice as much.Results Compared with IR group,70gene expression is different in the IR-P group. There are48genes were up-regulated and22genes were down-regulated. These genes mainly involved:Cell metabolism and damage,appoptosis-related protein,signal transduction and transcription, correlated inflammatory factor and oxidative stress related factors.Conclusion Ischemic postconditioning plays a role on multiple genes and acts on multiple targets to reduce ischemia reperfusion injury.
