This paper aims to evaluate the resistance of the DC cells from bone marrow of miceto mature characteristics and the relationship between the Jagged-2. Method between theuse of bone marrow mesenchymal stem cell co-culture and curcumin stimulus of twogroups of immature DC cell model is established, two groups of cells by flow cytometrytechnical appraisement of maturity, and use the LPS stimulation mature resistancecharacteristics of two groups of cells, and then by using flow cytometry and Rt-PCRtechnique in cell membrane protein level and the level of gene expression of Jagged-2inquantity for testing, the mouse paw inflammation model is set up in the body, the matureresistance characteristics of two groups of the experimental group in the body were studied.Results between bone marrow mesenchymal stem cells and DC cells co-culture andcurcumin stimulation can be successfully established in immature DC of cells, the cellsurface molecules of the two groups is similar to the immature DC cells, and two sets ofDC has the character of resistance to mature cells, after adding LPS stimulation, notexpress a similar mature DC surface molecules. Jagged-2in membrane protein levels inthe two groups have no obvious difference between experimental group and imDC group,and at the genetic level expression have significant differences, with mature resistancecharacteristics of two groups of experimental group is significantly higher than ordinaryimDC group. In the mouse paw inflammation model, the performance of the two groups ofexperimental group is superior to the ordinary imDC group, two groups of experimentalgroup can obviously inhibit the inflammatory response, and ordinary imDC group has nosuch effect. Conclusion:1, immature cells have immunosuppressive effect, bone marrowmesenchymal stem cells and curcumin can induce immature DC cells showed resistance tomature features.2, immature DC cells of mature characteristics and high expression ofJagged-2there is a close relationship.3, imDC group and two experimental groups inmembrane protein expression level no significant statistical differences in gene expression levels have significant statistical difference, it follows that may was statistically differencebetween intracellular protein levels, and decided to DC cells resistant to mature featuresmainly depends on the amount of protein expression intracellular Jagged-2.
