Objective:Aging is a complex process phenomena of life, there must be some changes in biomarkers of aging process, the paper aims to filter out age-related biomarkers, namely senescence markers, through its joint detection of biological Age discriminant analysis, using stepwise regression to establish biological age regression equation and observe its predictive value, and explore the relationship between age-related biomarkers in diseases associated with aging(type2diabetes and atherosclerosis).Methods:180cases of normal healthy people, Endocrinology,40patients with type2diabetes, Cardiology,40patients with atherosclerosis from First Affiliated Hospital of Soochow University.First part Detected180cases of normal healthy human plasma biomarkers (TIMP-1, IGF-1,IL-2,IL-6,CRP,TNF-α,Insulin) expression in Ⅰ、Ⅱ、Ⅲ、Ⅳ four groups, and the relationship with age by ELISA. The use of age-related biomarkers combined detection of biological age discriminant analysis using stepwise regression to establish biological age regression equation and observe its predictive value.Second part Detected the expression of age-related biomarkers in the plasma of patients with aging-related diseases (type2diabetes and atherosclerosis) by ELISA method to investigate the relationship between age-related biomarkers and aging-related diseases.Results:First part (1) TIMP-1, IGF-1, IL-2, IL-6, CRP,TNF-a and Insulin have significant difference among the four groups (P<0.05).(2) ELISA test results suggest that:TIMP-1, IGF-1, IL-2, IL-6, CRP have relationship with age in healthy human, the correlation coefficient was (r=0.3354,-0.5097, 一0.4940,0.4801,0.2859),P<0.05,statistically significant.(3)The use of age-related markers for discriminant analysis function to get six discriminant function analysis equation is:Y20-30=-27.369+85.689*TIMP一1+0.11*IGF一1+1.575*IL-2+0.68*IL-6-0.259*CRPY31-40=-27.654+76.445*TIMP一1+0.089*IGF一1+1.819*IL-2+0.848*IL-6-0.428*CRPY41-50=-27.446+83.857*TIMP一1+0.085*IGF一1+1.599*IL-2+1.233*IL-6-0.285*CRPY51-60=-25.466+104.809*TIMP一1+0.07*IGF一1+1.564*IL-2+1.002*IL-6-0.349*CRPY61-70=-22.653+116.683*TIMP一1+0.063*IGF一1+1.137*IL-2+1.242*IL-6-0.226CRPY>70=-22.925+135.232*TIMP一1+0.046*IGF一1+0.861*IL-2+1.506*IL-6-0.073*CRP(4)Stepwise regression analysis of biological age regression equation is: Y=61.313+147.643*TIMP-1-0.15*IGF-1-1.639*IL-2+1.641*IL-6(R2=0.594,F=38.923).And through residual analysis,regression model to determine the effect has been better,is reasonable.Second part(1)Plasma TIMP-1,IL-2,IL-6,CRP elevated compared with normal controls in type2diabetes and atherosclerosis patients,P<0.05,with statistical significance.(2)Plasma IGF-1lower than those in the normal control of type2diabetes and atherosclerosis patients,P<0.05,with statistical significance.Conclusion: With aging, a variety of physiological functions will be changes.TIMP-1,IGF-1,IL-2,IL-6,CRP have relationship with age and independent of each other, there is a certain relationship between age-related biological markers and age-related disease.Use these biomarkers to get the predicted biological age,it can be used for health care,health measures and the evaluation of the health of an individual on the basis of the same physiological age reference.
