Background and Objective:Endometriosis (EMs) is a common gynaecological disease of childbearing age women, often accompanied by pelvic pain and infertility, seriously affecting the quality of life and fertility of women. EMs is a benign disease, but has some similar tumor biological behavior such as proliferation, invasion and metastasis. The pathogenesis of EMs is not clear. With the advance of the pathogenesis research, gene regulatory role in the onset process is becoming a hot spot of research. NEK2in mitosis is mainly responsible for G2-M key point control, and promote the centrosome maturation and influence the concentration of chromosome and formation of spindle. Its abnormal expression was often associated with cell malignant transformation disease. It was also expressed excessively in a variety of tumor cells. On the other hand, it caused a split centrosome multipolar. This study has used molecular biological techniques to detect the expression of NEK2in women with endometriosis (ectopic group; eutopic group) and women without endometriosis. Preliminarily NEK2was associated with intrinsic connection of endometriosis disease development.Materials and Methods:All the tissues were retrieved from Women’s Hospital, School of Medicine, Zhejiang University, from March to May in2012. Ectopic (ectopic group, n=33) and eutopic endometrial tissues (eutopic group, n=19) were obtained from the women who did the ovarian cyst removal surgeries, and were confirmed histopathologically as endometriosis. Their Stage were III-IV and age were34±5. Normal endometrial tissues(control group,n=27) were collected from the women without endometriosis. Their age were33±7. All entrants had no physical or comorbid illness.They had regular menstrual cycles28-32d, and within3months before the operation they had not acceptted hormones and other drugs, nor intrauterine device. All collected fresh tissues samples were cleaned by ice PBS immediately before being put into liquid nitrogen and they were reserved for extracting mRNA and protein.Objects and Methods:A semiquantitative analysis of the expression of NEK2mRNA and protein in the endometrium was detected by RT-PCR and Western blotting in women with endometriosis (ectopic group,n=33; eutopic group,n=19) and women without endometriosis (control group, n=27). Experimental results used SPSS16.0software between3groups homogeneity of variance analysis and single factor variance analysis. Two groups compared with LSD-t test method, with P<0.05for the difference was statistically significant.Results:Both the expressions of NEK2mRNA and protein were significantly lower in ectopic endometrium than in eutopic and normal endometrium (P<0.01), while no difference was showed between the latter two groups (p>0.05).Conclusion:NEK2expressions are present in the endometrium, but down-regulated in ectopic group, suggesting that NEK2may be involved in cell cycle regulation in the development of endometriosis.
