Objective A case-control study to explore the validation VEGFR-2gene promoterpolymorphism is associated with atherosclerosis risk of ischemic stroke and VEGFR-2gene promoter region polymorphism and CISS classification under atherosclerosispathogenesis of cerebral infarction subtypes whether each has its relevance.Methods According to case-control study to collect150cases.Due to blood loss orother reasons, the final case group are103patients and the control group are43patients.Cases were CISS artery atherosclerosis in patients with ischemic stroke, according to itspathogenesis is divided into four subgroups; the control group are without symptoms ofstroke and non-stroke patients with mild.Patients with peripheral venous blood drawn and extraction of whole blood DNA. Weselect VEGFR-2gene promoter-604T/C locus by PCR-restriction method for genotyping.After multivariate Logistic regression model adjusted for conventional cardiovasculardisease risk factors, analysis of gene polymorphism in the promoter region of the relevanceof the risk of stroke and pathogenesis of various subtypes of the risk groups and theircorrelation.Results VEGFR-2gene promoter region-604C allele frequency of case group issignificantly lower than the control group (9.7%in the case group and control group45.3%,P <0.0001) and-604TC and CC genotype frequencies in case group was significantlylower than the control group (case group TC12.6%CC3.9%, control group, TC44.2%CC23.3%, P<0.0001). Subgroup analyzes found there is no statistically significant interms of genotypes between subgroups (P>0.05), Allele C frequencies are lower than thecontrol group in subgroups Artery-to-artery embolism, Parent artery thrombus occludingpenetrating artery and Multiple mechanism (P values <0.05).-604TC and CC frequency isalso lower than the control group (P values <0.05). Subgroup Hypoperfusion impairedemboli clearance allele frequencies and genotype frequencies of-604TC and CC, although lower than it in the control group, but with the corrected chi-square test it shows P values>0.05. Logistic regression analysis after correction with the related risk factors andprotective factors, VEGFR-2gene promoter region-604C allele is still associated withdecreased risk of ischemic stroke-related.Conclusion VEGFR-2gene is susceptibility gene for atheroscleros ischemic stroke;VEGFR-2gene promoter region-604C allele was associated with reduced atheroscleroticischemic stroke risk, in which the main arteries CISS typing-artery embolization,perforating artery occlusion and mixed mouth subtype of the more obvious.
