Objective:By building a model of non-alcoholic fatty liver in vitro,study itseffects on related inflammatory factor expression and its activation of UPR way inliver cells. To further explore the possible molecular mechanism of liver cell injurythat causes by high-fat emulsion. And to observe whether MgIG(magnesium isoglycyrrhizinate) has a protective effect on the model,to provide clinical experiment basis of MgIG used in the treatment of NAFLD(non-alcoholic fatty liver disease).Methods: Building a model of NAFLD,to observe and evaluate the construction rate and screen the best fat emulsion concentration and incubation time with thesurvival curve,oil red O staining and light microscope. Incubate nonalcoholic fattyliver cells with1mg/ml MgIG. Detect the expression on mRNA level of IL-6,IGF-1,TGF-β,bFGF,BIP,XBP-1and CHOP with Real-time PCR.Results:(1)To the cell,the function of fat emulsion in certain concentration ispromoting pimelosis,but it has cytotoxin effect with the increase of the concentration,the best concentration is0.1%and0.2%,the incubation time is24h and48h;(2)In model,the level of IL-6,IGF-1,TGF-β,bFGF expression on mRNA showa trend of rising;(3) The related factor expression of MgIG control is reduced.Conclusion:1.the fat emulsion can be used to build in vitro nonalcoholic fattyliver models;2. it may activate molecular nf-kb to make more inflammatory factors of liver cells release,which can cause liver cells injured;3.MgIG has certain protective effect on high fat emulsion load liver cells.
