Object:To investigate the inflammation regulation function of human placenta derived mesenchymal stem cells in OA mediate acute lung injury;and detected in injury lung tissue, detected the expression of RhoA after transplation hpMSCs with the acute lung injury rats. Further our reacher will reveal hpMSCs how to regulate the inflammation in acute lung injury, discuss the potentiality of cellular therapy of acute lung injury.Method:1. hpMSCs were isolated from full-term pregnancy placenta and cultured by collagenaseⅣ. The enzyme digestion can effectively isolate placenta mesenchymal stem cells with a high proliferative ability and multipotent differentiation ability. To understand the general characteristics by the in vitro culture, cell counts, to nerve, bone induction.2. Acute lung injury model was establish by injecting Oleic acid (OA) into sublingual vein(0.2ml/kg).Briefly,36male SD rats were randomized into three groups:1) acute lung injury plus hpMSCs administration, n=12.2) acute lung injury plus normal saline n=12;3) normal control n=12.Animal were killed at24h after OA exposure respectively. Blood samples were collected to detect plasma IL-8and IL-10expression using ELISA. Lungs were removed and used for evaluation of wet/dry ration.Part of the lung tissues were paraffin-embedded to assess lung tissue damage quantitatively and detect the expression of RhoA of lung tissue in different group by RT-PCR and western-blot,Result:1) The mult idiferention of hPMSCs, the reproductive activity of hpMSCs was attenuated after passages. The in vitro. inductive culture demonstrates cartilage and neural differentiation. 2) Role of pMSCs transplantation in acute lung injury:L+M group showed a obvious amelioration in tissue injury when compared with group L compared with the normal saline group, the W/D, IL-8level in plasma, the expression of RhoA were significantly increased. IL-10in plasma level were decreased.But in hpMSCs treatment group these indexs were siginificantly lower than those in the ALI group.Conclusion:1) The hpMSCs isolation and purification were successful by collagenase IVdigestion and stem cells obtained were multipotency.2) The hpMSCs transplation in early phase of ALI reveal cell morphological changes. And the W/D value, plasma IL-8level was degraded; plasma IL-10level was upgraded.suggesting benefits to tissue repair and inflammation resolution.3) The level of RhoA was increased in the actue lung injury,hpMSC transplation may decrease RhoA expression. We could conclusion that hpMSC transplation could balance inflammatory factor expression in the acute lung injury by regulation RhoA...
