Aim: Screen for miRNAs of plasma which are abnormally expressed in rheumatic heartdisease with pulmonary hypertension, get miR-1183and miR-1299two different biomarkers, andmake a preliminary study as miR-1183and miR-1299as two different biomarkers in expandingsample of rheumatic heart disease with pulmonary hypertension.Methods: By extracting rheumatic heart disease in patients with pulmonary hypertension andnormal RNA in plasma, and using the gene microarray to detect the miRNA expression profiling inplasma of rheumatic heart disease with pulmonary hypertension. According to the gene chip results,select miR-1183and miR-1299as a research object, using qRT-PCR to test expression differencebetween in rheumatic heart disease with pulmonary hypertension and normal human.Results: Chip results suggest that miR-1299, miR-1183, miR-4455, miR-3148, miR-4660,miR-3149, miR-4682etc. which expressed in rheumatic heart disease with pulmonaryhypertension were significantly upregulated; whereas miR-423-5p, miR-425-5p, miR-744-5p,miR-208b, miR-326, miR-636etc. expressed in rheumatic heart disease with pulmonaryhypertension were significantly downregulated. Therefore, the expressions of miR-1183and miR-1299in rheumatic heart disease with pulmonary hypertension were detected by qPCR, and theresult was consistant with what the miRNA microarray represents. According to SPSS softwareanalysis, the expression of miR-1183in rheumatic heart disease with pulmonary hypertension wasupregulated(p=0.012), meanwhile, the expression of miR-1183in severe pulmonary hypertensionwas upregulated(p=0.021); the expression of miR-1299in rheumatic heart disease with pulmonaryhypertension was upregulated(p=0.011), but there is no significant difference on the expression ofmiR-1299between in severe pulmonary hypertension and mild pulmonary hypertension (p=0.566).Conclusion: The miRNA of plasma was differentially expression between in rheumatic heartdisease with pulmonary hypertension and normal and miRNA might be independent effect inrheumatic heart disease and pulmonary hypertension, thus illustrate not only the miRNA asbiomarkers significance for rheumatic heart disease, but also pulmonary hypertension has somerelevance.
