ObjectiveWe studied the methylation of septin9promoter in cervical cancer and stomach cancer, andobserved the change of septin9expression in cervical cancer and stomach cancer cell lines aftertreating with SAMe. And then discussed the significance of septin9promoter methylation incervical cancer and stomach cancer, and the possible mechanism which SAMe induce the tumorcells apoptosis.MethodsFirst, the methylation sensitive high resolution melting curve analysis (MS-HRM) technologywas used to detect the methylation of septin9promoter in cervical cancer cell line HeLa, gastriccancer cell line MGC and cervical cancer tumor tissues; and analyzed the relationship between themethylation of septin9promoter and cervical cancer tumor tissue clinical pathology process.Second, we used DAPI nuclear staining to observe apoptosis of cervical cancer cell line HeLa andgastric cancer cell line MGC which treated24h,48h by SAMe. Finally, through the quantitativereverse transcriptase-polymerase chain reaction (qRT-PCR) method and MS-HRM technology, wedetected the methylation of septin9promoter and mRNA expression level of septin9-v1,v5,v inSAMe-treated cervical cancer cell line HeLa and gastric cancer cell line MGC.Results1. Aberrant methylation of septin9promoter was existed in cervical cancer cell line HeLa andgastric cell line MGC.2.38.5%(5/13) of cervical tumor tissues detection showed septin9promoter aberrantmethylation, but no significant correlation of septin9promoter aberrant methylation was observedin clinical staging of cervical cancer, lymph node metastasis and the presence or absence of bloodvessel invasion (P>0.05).3. We found that HeLa cervical cancer cell line and MGC gastric cancer cell line appearedapoptosis by SAMe treated24,48h.4. The degree of the methylation of septin9promoter in SAMe-treated cell lines was similarwith the control groups. 5. Compared with the control groups, mRNA expression level of septin9-v1,v5,v werereduced significantly in HeLa cervical cancer cell line and MGC gastric cancer cell line of treatedgroups.ConclusionsThe study showed the aberrant methylation of septin9promoter was existed in cervicalcancer and stomach cancer. Although SAMe could not reverse the methylation sites of septin9, butseptin9-v1mRNA expression in SAMe-treated cervical cancer and gastric cancer cell lines weresignificantly decreased. And found that cervical cancer and stomach cancer occurred the apoptosis.We inferred that SAMe possibly acted on septin9by a certain way to decrease the expression ofseptin9-v1in cervical cancer and stomach cancer, then hindered JNK signaling pathway whichinvolved in cell proliferation, thus resulted in apoptosis of cervical cancer and gastric cancer celllines.
