The Study Of The Effects And Mechanisms Of Adiponectin On Podocytes Apoptosis

Object：The purpose of this project is to study under conditions of high glucose orPAN, the apoptosis rate of podocytes with or without the adiponectin（2.5ug/ml）adiponectin.Method：The podocytes were treated with normal glucose （NG,5.5mmol/L）, highglucose（HG,30mmol/L）, HG with gAd（2.5ug/ml）, PAN（50mg/L）and PANwith gAd（2.5ug/ml） respectively. The apoptosis rates of podocytes with differenttreatments were detected using AnnexinV/PI and in situ end labeling (TUNEL)methods after12h,24h,48h. The apoptosis-related genes Bax, Bcl-2and caspase3mRNA levels were detected by realtimePCR. Finally, the levels of apoptosis-relatedproteins Bax, Bcl-2, caspase3expression were measured using western blotting.Results：(1) The apoptosis rates of podocytes in HG group were higher than the NGgroup and lower than HG with gAd group. The apoptosis rates in PAN group werehigher than the NG group and lower than PAN with gAd group.(2) The levels ofBax and caspase-3mRNA in HG group were higher than NG group and HG withgAd group. Caspase3and Bax mRNA levels in PAN group were higher thanNG group and PAN with gAd group also.(3) Western-blot detect the expression ofapoptosis related proteins Bax, Caspase3：The levels of apoptosis-regulated proteinssuch as Bax and caspase-3in HG group were higher than NG group and HG withgAd group，and their levels in PAN group were higher than NG group and PANwith gAd group.Conclusions：The apoptosis rates of podocytes treated with HG or PAN wereattenuated after incubating with adiponectin. The levels of Bax bcl-2andCaspase3mRNA were decreased.The levels of apoptosis-regulated proteins such asBax and caspase-3in HG group were higher than NG group and HG with gAdgroup，and their levels in PAN group were higher than NG group and PAN with gAdgroup. All the results show that adiponectin can inhibit the apoptosis of podocytes。...
