Objective: To observe the effect of HBx gene on the proliferation of HL-7702cells,and investigate the effect of IL-10on the proliferation of HL-7702cells stablyexpressing HBx gene and the interaction of the expression of CDKN1B mRNA.Methods: HL-7702cells, HL-7702/MOCK cells and HL-7702/HBx cells werecultured in vitro cell culture. The effect of HBx gene on the proliferation ofHL-7702cells and the effect of IL-10on the proliferation of HL-7702cells stablyexpressing HBx gene were detected by CCK8assay. Flow cytometry was used toanalyze the effect of IL-10on the apoptosis of HL-7702/HBx cells. RT-PCR wasused to detect the effect of HBx gene on the expression of CDKN1B mRNA inHL-7702cells and the effect of IL-10on the expression of CDKN1B mRNA inHL-7702/HBx cells.Result: The data of CCK8assay showed that the proliferation rate of HL-7702/HBxcells was significantly faster than HL-7702and HL-7702/MOCK cells, and therewas no difference between the proliferation rate of HL-7702cells andHL-7702/MOCK cells. In current study, we found that after24hours,80ng/ml IL-10have significantly inhibited the proliferation of the HL-7702/HBx cells, howeverthere was no effect on the proliferation of HL-7702and HL-7702/MOCK cells aftertreatment with IL-10. In addition, there was no effect on the apoptosis of HL-7702,HL-7702/MOCK and HL-7702/HBx cells after24hours exposed to80ng/ml IL-10.Compared with HL-7702and HL-7702/MOCK cells, the expression of CDKN1BmRNA level in HL-7702/HBx cells had decreased, and there was no differencebetween the expression of CDKN1B mRNA level of HL-7702cells andHL-7702/MOCK cells. We also found that treatment with80ng/ml IL-10for24hours had increased the expression of CDKN1B mRNA level in HL-7702/HBx cells.Conclusions: HBx gene can promote the proliferation of HL-7702cells. IL-10caninhibit the proliferation of HL-7702/HBx cells but have no effect on the apoptosis. HBx gene probably enhanced the proliferation of HL-7702/HBx cells bydown-regulating the expression of CDKN1B mRNA. IL-10can inhibit theproliferation of HL-7702/HBx cells which was probably related to the up-regulationof the expression of CDKN1B mRNA.
