ObjectiveTo study the function of Sonic hedgehog(Shh) in maternal serum in theplacental of women with preeclampsia(PE).MethodsA total of60women with PE and60normal pregnant women as controlparticipated in this study.All were admitted to Fujian Maternity and Child HealthHospital for delivery from Octomber2012to December2013.Patients with PE weredivided into early-onset group(n=30,presented at<34weeks of gestation) andlate-onset group(n=30,presented at≥34weeks of gestation),with30normalpregnant women as early control group(<34weeks of gestation) and30as latecontrol group(≥34weeks of gestation).Enzyme-linked immunoassay(ELISA) was used to detect Shh in maternalserum. Immunohistochemistry was used to detect the location of Shh expression inplacenta. Western blot was used to detect Shh proteins expression in placenta.Real-time fluorescent quantitative PCR was used to detect Shh mRNA expression inplacenta.Results①No significant difference was found in the mean level of Shh in maternal serumbetween the early-onset and late-onset preeclampsia groups with the controlgroups(P>0.05).②Protein of Shh was expressed in placenta of all groups. Trophoblast cellscontained brown granules,with the majority in the cytoplasm. ③Western blotting revealed that the level of placental Shh in the early-onset andlate-onset preeclampsia groups was significantly higher than in normal placenta(P<0.05).④The level of placental Shh mRNA was significantly elevated in the early-onsetPE group and late-onset PE group as compared to late control group(P<0.05).ConclusionShh is likely to be involved in preeclampsia through some common mechanism. ObjectiveTo study the influence of different concentrations of3-hydroxy palmitic(3-HPA)oxidative stress damage on placenta tissues.MethodsWomen with PE and normal pregnant was admitted to Fujian Maternity andChild Health Hospital.Placenta tissues from normal pregnancy women was incubatedwith normal, preeclampsia serum and different concentrations of3-hydroxypalmiticfor24hours.Ultraviolet colorimetry was used to detect malondialdebyde (MDA).Totalsuperoxide dismutase (SOD) assay kit-WST was used to detect SOD activity inplacenta.The fluorescent dye assay was applied to measure the mitochondrialmembrane potential.Results①The levels of MDA in3-HPA and preelampsia groups were higher than theircontrol groups,the differences were statistically significant(P<0.05).②The levels of SOD activity in3-HPA and preelampsia groups were lower than their control groups,the differences were statistically significant(P<0.05).③Through the analysis we found that the higher cholesterol concentrations(0,25,50,100μM) in medium, the higher oxidative stress damage(P<0.05). Themean placental MDA concentration correlated negatively with placentalantioxidant enzyme in the early-onset PE group and late-onset PE group(P<0.05).ConclusionsHigh levels of oxidative damage in placental mitochondria may be involved inthe pathogenesis of preeclampsia. Increased3-HPA and decreased activity ofantioxidant enzyme in maternal serum may contribute to high oxidative damage levelsin placenta in women with preeclampsia. ObjectiveTo investigate preeclampsia serum,Shh and3-HPA on the influence of oxidativestress damage and cell apoptosis of trophoblast cell.MethodsCulture trophoblasts cells(HPT-8) in the medium with normal pregnancyserum,preeclampsia serum,3-HPA,preeclampsia serum with recombined Shh andpure medium for24hour.Ultraviolet colorimetry was used to detect malondialdebyde(MDA).Totalsuperoxide dismutase(SOD)assay kit-WST was used to detect SOD activity in cells.Annexin V/PI method were used to detect different cell necrosis and apoptosis.Results①The levels of cell oxidative stress damage and apoptosis in pre-ecampsia group were higher than normal pregnant group,the differences were statistically(P<0.05).②The levels of cell oxidative stress damage and apoptosis in3-HPA andpreelampsia groups were higher than their control groups,the differences werestatistically significant(P<0.05).③The levels of cell oxidative stress damage and apoptosis in preeclampsia serumwith recombined Shh group were lower pre-ecampsia group than the differenceswere statistically(P<0.05).ConclusionsThe preeclampsia serum can lead to trophoblast cell oxidative stress damage cellapoptosis.Long chain fatty acids may be involved in the pathogenesis of preeclampsiapossible through the enhancement of oxidative stress damage and cell apoptosis.Shhprotective effect on cell oxidative stress damage cell apoptosis.
