Objective: To investigate the expression of Shh(Sonic Hedgehog)andGli2(Glioma-associated oncogene homolog2) in laryngeal squamous cell carcinomaand analyze their clinical significance and the relationship between Shh and Gli2.Methods:1.The expression of proteins of Shh and Gli2was analyzed byStreptAvidin-Biotin-enzyme Complex(SABC) Immunohistochemistry staining in60cases of laryngeal squamous cell carcinoma.30cases of atypical hyperplasia oflaryngeal and30cases of were studied as controls.2.The expression mRNA of Shh and Gli2were analyzed by reversetranscription-polymerase chain reaction(Rt-PCR) in50cases of laryngeal squamouscell carcinoma,40cases of the cancer cut margin were studied as controls.Andclinical significance and the their relationship with mRNAs were also be counted.3.The expression proteins of Shh and Gli2were analyzed by Western Blot in50cases of laryngeal squamous cell carcinoma,40cases of the cancer cut margin werestudied as controls.And clinical significance and the their relationship with proteinswere also be counted.Results:(1)The result of Immunohistochemistry showed:a、The positive ratesof Shh protein were10.00%(3/30)、33.33%(10/30)、60.00%(36/60) in laryngealpolyp、 atypical hyperplasia of laryngeal、laryngeal carcinoma, and that of Gli2protein were6.67%(2/30)、23.33%(7/30)、45.00%(27/60). Three groups werestatistically significant(P<0.05).b、The positive rates of Shh was addition when theclinical stage was higher. The positive rates of Shh in clinical stage of I+II、III+IVwere44.45%、83.33%.And the rate of Gli2were33.33%、62.50%. Two groups werestatistically significant(P<0.05).c、The positive rates of proteins of Shh、Gli2were addition when the histological grade was higher.The positive rates of protein of Shh inwell differentiated group and the moderately-poorly differentiated group respectivelywere41.18%、84.62%.And the rates of protein of Gli2were32.35%、61.54%.Comparing the two groups of the two proteins were statistically difference(P<0.05).d、The positive rate of Shh protein in the group of lymph node metastasis was88.24%.It was apparently higher than no lymph node metastasis group. Comparing thetwo groups was statistically difference(P<0.05).Protein of Gli2in the two groups hadthe same tendency,and the positive rates were64.71%、37.21%.It also had thestatistically difference(P<0.05).e、The expression of proteins of Shh and Gli2were notrelated to patients’ gender、age、alcohol、smooking or tumor place(P>0.05).(2) Theresult of Rt-PCR show:In50cases of laryngeal squamous cell carcinoma,40cases ofthe cancer cut margin tissue the positive rates of Shh and Gli2mRNA in laryngealsquamous cell carcinoma were much higher than in cut margin of the cancertissue(Shh:64.00%、25.00%,P<0.05;Gli2:48.00%、20.00%,P<0.05).Theexpression of Shh and Gli2mRNA in laryngeal squamous cell carcinoma weresignificant difference than in cut margin of the cancer tissue(Shh: P <0.05;Gli2: P<0.05).(3)The result of Western Blot show:In50cases of laryngeal squamous cellcarcinoma,40cases of the cancer cut margin tissue the positive rates of Shh and Gli2proteins in laryngeal squamous cell carcinoma were significant difference than in cutmargin of the cancer tissue(Shh, P <0.01;Gli2, P <0.01).The expression of Shh andGli2proteins were related to histological grade、clinical stages、pathologic stages andlymph node metastasis(P <0.05), but not related to patients’ gender、age、alcohol、smooking or tumor place (P>0.05).(4) Shh protein and Gli2protein have positivecorrelation,so do their mRNA expressions (IHC: r=0.546, P <0.05;Rt-PCR: r=0.59,P <0.05; Western blot: r=0.61, P <0.05)Conclusion:(1)Maybe the abnormal expression of Shh、Gli2involved in laryngealsquamous cell carcinoma development、invasion、metastasis.(2)Sonic Hedeghog signaling pathways may be some effect in laryngeal squamouscell carcinomas happening and development. (3)Combined detection the expression of Shh、Gli2as biological markers can helpdiagnosis and treatment in laryngeal squamous cell carcinoma...
