Objective The mouse keratocytes were infected by Fusarium Solani to observethe molecular changes in them. Otherwise, FA、FB1、DON、ZEA were studied for theexpression and potentional role in fungal keratitis.Methods Mouse keratocytes were subcultured by the method of enzymedigestion, and a standardized inoculum (107cFu) of Fusarium Solani was culturedwith keratocytes. On day1,2after inoculation, the molecular changes was observedby micro-Raman spectroscopy using near-infrared laser (514um) excitation;Physiological saline was regarded blank control group. At1、3、5d after inoculation,LC-MS/MS was detected for the expression of FA、FB1、DON、ZEA.Results In the study, the location and shape of the average Raman spectrum ofthe infected keratocytes was similar to the ones normal, but the intensity of somecharacteristic peaks decreased, reflected at1005cm-1、1186cm-1、1657cm-1、1449cm-1、and1311cm-1, respectively belong to phenylalanine、tyrosine、α-helix、membrane phospholipids and nucleic acid. The result of LC/MS method showed thatFB1and DON can be detected from the cell culture.Conclusions The Fusarium Solani can inhibit the protein synthesis、change themembrane structure and damage the genetic material, leading to the cell die. The FB1and DON can be detected in the infected group at5d. Mycotoxins may play asignificant role in the process of the fungal keratitis.
