Unraveling The Effect Of Hypoxic Preconditioning In Focal Cerebral Infarcted Rat Neurons Ion Channel Gene Expression By RT~2Profiler Pcr Array

Objective To analysis the Neuronal ion channel gene expression in hypoxiapreconditioning and PMCAO rats,to discuss the effect and molecular of Neuronal ionchannel gene subunits in ischemic injury.Methods1. Sixty-four SD-rats were randomly divided into five groups, control group (GroupCB, n=8), sham-operated group (Group NF, n=8), the operated of middle cerebralartery occlusion group (Group NM, n=20), hypoxia-preconditioned group(Group HB,n=16), hypoxia-preconditioned and middle cerebral artery occlusion group (GroupHM, n=20). The NM, HM,HB group were divided into6hours and24hours afterpermanent middle cerebral artery occlusion each subgroup have ten rats.2. Rats in hypoxia-preconditioned and hypoxia-preconditioned+PMCAO group wereplay in simulated altitude of5000m in the low pressure oxygen tank, two hours a day,at the same time for three consecutive days.3. Rats in hypoxia-preconditioned+PMCAO and PMCAO group were made MCAOmodel by Zea longa method.4. To evaluate the rats nerve function score by Longa5points method after PMCAO6hours and24hours.5. The ischemic region was visualized by2%TTC staining method and calculatecerebral infarction area.6. To analysis neuronal ion channels in rats by RT2Profiler PCR Array.Results1. Under the treatment of hypoxia-preconditioned, In6h and24h time point,theneurological impairment in HP+MCAO group was alleviated than MCAO group.(p<0.05).2. In6h and24h time point, the infarct volume HP+MCAO group was less thanMCAO group.(p<0.05).3. RT2PCR Arrays results revealed29cases of gene up-regulated as comparedMCAO group with HP+MCAO group at6hours after PMCAO,Which rise in Kcnj15is most obvious, increase ratio of17.1051.11cases of gene down-regulated and1cases of gene up-regulated as compared MCAO group with HP+MCAO group at24hours after PMCAO, the up-regulated genes is Trpc3.Which Cacng4and Kcnj4difference is obvious in down-regulated gene. Conclusions1. Hypoxia preconditioning alleviated the neurological impairment and reduced theinfarct volume, which also reduce the damage of neurons. Our results provideevidence indicating that Hypoxia preconditioning has important significance ofprotect neurons.2. The RT2Profiler PCR Array results reveled that the differentially ion channel genesmay be involved in hypoxia preconditioning protective, Further research will continueto help to unravel the molecular mechanism.
