Objective:We used ion exchage chromatography and affinity chromatography methodspurifying NanC protein. To investigate the effects of NanC protein on cellproliferation and apoptosis in human acute promyelocytic leukemia cell line HL60and human acute mononuclear leukemia cell line THP-1.Methods:(1). NanC protein was tested by SDS-PAGE electrophoresis.(2)To assay demonstrated concentrations of NanC protein in cell proliferation,cell cycle and apoptosis.(3). Using realtime-PCR assay and western blot assay to examine mRNA leveland protein level of genes, and to research the mechanism of NanC protein in HL60and THP-1cells in vitro.(4).Western blot assay showed the main cell signal of NanC in cancer cells invitro.Results:(1). Results of SDS-PAGE showed that the purity of NanC protein was higher.(2). NanC protein showed valid anti-cancer effects in vitro. NanC proteininhibited cell proliferation and induced apoptosis.(3). Results showed that NanC induced apoptosis by activating apoptosis-relatedgenes Bax and Bcl-2.(4). NanC could activate cell signal NF-κB and regulated the expressions ofp65gene gene.Conclusion:(1).NanC protein demonstrated a range of effects in human cancers, and itinhibited cell proliferation, induced apoptosis by regulating classic cell signals invitro.(2). The effects of NanC in vitro were associated with NF-κB signal and NanCregulated family members of NF-κB to regulate gene expressions.
