Objective:To explore the role of HSP60in diabetic cataract by observe the expression andchange of HSP60in lens epithelial cells in early diabetic rats;Observe the role ofHSP60on lens epithelial cell apoptosis.Methods:6-8weeks SD rats were divided into normal control group and diabetes group.l%STZ were intraperitoneal injected to induce diabetic model in experimental group,4,8,12weeks respectively after STZ-induced, the expression and the change of thequantity of HSP60in lens epithelial cells were analyzed by immunohistochemical andWestern blot in normal control rats and diabetic rats. Lens epithelial cells line(SRA01/04) were cultured in DMEM medium containing20%fetal bovine serum and5mmol/L glucose, then the cells were divided into two groups and were cultured inhigh concentration glucose (25mmol/L), one group of HSP60protein (10mg/L) wereadd to the medium, the another group as the control, after72h the analysis ofcaspase–3by Western blot.Results:There are no HSP60expression in lens epithelial cells in normal rats. At4w、8w and12w after STZ-injected, there are HSP60expression in lens epithelial cells indiabetic rats.The amount of expression began to decrease at12W after STZ-injected,there are significant differences (P <0.05).but the expressing quantity there are nosignificant difference between4w and8w week (P>0.05). Caspase-3expressionin lens epithelial cells were decreased in HSP60protein group.Conclusion:There are a great number of HSP60expression in lens epithelial cells in the earlystage of diabetic rats, but with the extension of time, the expression quantity willdecline.HSP60has a protective role on the lens epithelial cells apoptosis induced byhigh glucose.
