Objective: To investigate the effects of Sinomenium on the nf-kappa B andcyclooxygenase-2expression in the periaqueductal grey of the rat migraine model, and toenrich the mechanism of action of sinomenine treatment of migraine.Methods:60healthy Sprague-Dawley rats (half female and half male) were randomlydivided into blank group (10rats) and model group (50rats). The model group wasestablished by subcutaneous administration of nitroglycerinl (10mg/kg/d) in the back oftheir necks,1time2days for5times. The blank group was injected subcutaneously by NS(10ml/kg/d),1time2days for5times. After the success of the migraine model making,rats were divided into NS comparison group,nimesulide treatment group, Sinomenium lowdose treatment group, Sinomenium middle dose treatment group, and Sinomenium highdose treatment group. From the second subcutaneous administration of nitroglycerinl30minutes later, the NS comparison group was fed with NS (20ml/kg/d) every day, thenimesulide treatment group was fed with nimesulide (6mg/kg/d) every day, theSinomenium low dose treatment group was fed with Sinomenium (50mg/kg/d) every day,the Sinomenium middle dose treatment group was fed with Sinomenium (100mg/kg/d)every day, and the Sinomenium high dose treatment group was fed with Sinomenium(200mg/kg/d) every day. After the last treatment of3hours, the positive cells of thenf-kappa B and cyclooxygenase-2in the periaqueductal grey area of the migraine rats wereevaluated by immunohistochemistry method.Results: The model group and treatment group appeared some special behavior andsymptom such as red ears, scratching head and climbing hutch about30minutes aftermodeling. The blank group did not appear the behavior and symptom. In contrast to NScomparison group, the time of turning red of eas and scratching head of the Sinomeniummiddle dose treatment group and high dose treatment and nimesulide treatment group rats disappeared which was significantly reduced. The frequency that the treatment groupsscratched their heads was significantly less than NS comparison group in different timespan. In contrast to blank group, the nf-kappa B and cyclooxygenase-2expression in theperiaqueductal greys within model group increased significantly, the difference wasstatistically(P<0.05). In contrast to NS comparison group, the nf-kappa B andcyclooxygenase-2expression in the periaqueductal greys within Sinomenium high-dosetreatment group and nimesulide treatment group decreased significantly, the difference wasstatistically (P<0.05). In contrast to nimesulide group, the nf-kappa B andcyclooxygenase-2expression in the periaqueductal greys within Sinomenium high-dosetreatment group, was not statistically difference(P>0.05).Conclusion: The up-regulation of nf-kappa B and cyclooxygenase-2expression in theperiaqueductal grey may be related to the outbreak of acute migraine. The Sinomenium iseffective in relieving the behavior and symptom of nitroglycerin induced hyperalgesia inmodel rats of migraine, one of the mechanism maybe prevent the nf-kappa B andcyclooxygenase-2expression in the periaqueductal grey.
