Preparation Of Immune Adsorbent For Rheumatoid Factor And Studies On Propert Of Adsorption

Rheumatoid arthritis is a chronic systemic autoimmune disease characterized byathrosynovitis, during which produce large amounts of rheumatoid factor and immunecomplexes that induce autoimmune inflammation. Hospitals mainly use drug-basedtreatment supplemented by perfusion. The purpose of this article is to preparerheumatoid factors adsorbent with high adsorption capacity, excellent selectivity andgood blood compatibility.Principle: Rheumatoid factor positive charge and has strong hydrophobic underphysiological conditions. The adsorbent connected phenylalanine to heparin hascarboxyl group(-COOH), It can adsorb RFs through ionic bond and hydrophobicinteraction.Methods: PS-Cl acts as carrier, which is connected with the linker heparin(HEP),then we connect phenylalanine to heparin(PHE). Researched the adsorption rate ofadsorbent by in vitro RF dynamic perfusion. We conduct in vitro elution experiment tomeasure the abscission rate of RF. Selectivity of adsorbent was evaluated by the bloodcomponents before and after perfusion. The prothrombin time(PTs) and thrombintime(TTs) detection in vitro verify the blood compatibility of adsorbent.Results:(1)The optimal condition of heparin connection: Initial concentration ofheparin is6mg/ml, lasting24h. And the grafted degree is33.8μg/g.(2)The optimalcondition of phenylalanine: Initial concentration of phenylalanine is6mg/ml, lasting24h. And the grafted degree t is8μg/g.(3) The adsorption equilibrium can be reachedin less than two hours, and90%of the maximum adsorption capacity was reached onlyunder1hour, while the adsorption rates of IgA RF、IgG RF and IgM RF in serumwere45%1.80、56%1.36、52%2.01(n=5). After elution, the abscission rate of IgARF、IgG RF and IgM RF were22%1.65、19%1.06、21%1.35(n=5).(4) Afterperfusion experiment, The variation rates of albumen and red blood cell were less than10%.(5) The detection results of PTs and TTs shows PS-PHE can delay the rare ofcoagulation.Conclusion: PS-PHE Using PS-Cl as carrier, HEP as connecting arm connect withthe linker PHE has higher adsorption rate to RFs, good selectivity ability and showsexcellent blood compatibility. PHE has a good research value and application prospect in treatment of rheumatoid arthritis.
