Objective: To search the effects of PUFAs (different ratio of ω-6PUFAs andω-3PUFAs)on the secretion of MMP-13and related mechanism at the cellular level.Methods: Chondrosarcoma cells (SW1353) were cultured in dulbecco’s modifiedeagle’s medium(DMEM)a period of time and then stimulated with inflammatorycytokine(IL-1). The cells were given chemicals in various mixture ratios(1~10) ofω-6PUFAs and ω-3PUFAs. Western blotting was used to quantify the expression ofMMP-13presence or absence of different mixture ratios of ω-6PUFAs andω-3PUFAs.Results: Supplementation with PUFAs reduced IL-1-induced release of MMP-13from sw1353explants without affecting the cells’ proliferation. The ratio ofω-6PUFAs and ω-3PUFAs(1~10)significantly inhibiting the the expression ofMMP-13is6:1.Conclusion: The optimum mixture ratio of ω-6PUFAs and ω-3PUFAs inhibiting thethe expression of MMP-13in articular chondrocytes of osteoarthritis is about6:1.
