ObjectivesResearch somatostatin analogue octreotide and DNA nucleotide analoguegemcitabine combined with human pancreatic cancer cells Capan-1proliferationexplore the combined effect between the two drugsexperimental and theoretical basisfor the treatment of pancreatic cancer drug.MethodsHuman pancreatic cancer cells Capan-1as experimental subjects, divided into fourgroups, the octreotide group, gemcitabine gemcitabine group and the combinationgroup.CCK-8assay analysis of different concentrations of octreotide withgemcitabine alone and in combination Capan-1proliferation of human pancreaticcancer cells; effect principle analysis of the effects of the two drugs in combination.Results1.Octreotide (1×10-6-1×10-1umol/L) and gemcitabine (3×10-8-3×10-3umol/L)alone and the combination (octreotide1×10-6-1×10-1umol/L+gemcitabine3×10-8-3×10-3umol/L) inhibited human pancreatic cancer cells Capan-1growth andproliferation of cells of the combined treatment group compared with the controlgroup and the monotherapy group was significantly reduced relative proliferativecapacity and dose-dependent manner, and the difference was statistically significant(P <0.05). 2. Effect principle analysis of the combination of the two drugs effect: theoctreotide effect concentration (Dm)5.2836×10-3umol/L, the gemcitabine in effectconcentration (Dm)3.6543×10-5umol/L, the combination effective concentration(Dm) of3.0154×10-4umol/L, the dose of the combination group was significantlylower than the individual dose. Effect principle analysis prompted the two drugs incombination have a synergistic effect (CI <1).Conclusion1.Octreotide with gemcitabine alone and in combination are able to inhibit thegrowth and proliferation of pancreatic cancer cells Capan-1cells.2. Octreotide combination with gemcitabine cells relative proliferation thanoctreotide with gemcitabine alone significantly reduced, in effect principle, the twodrugs combined have a synergistic effect.
