Objective:This study is using molecular targeted drugs sorafenib treat Hela cells,to observe the inhibition of sorafenib on the growth of Hela cells, and to study theinhibition of sorafenib combined with cisplatin, paclitaxel on the proliferation of Helacells, To explore the effect of sorafenib impact on the expression of VEGFR-3andPDGFR-β in cervical cancer Hela cells from molecular mechanism, provide atheoretical basis for the biological treatment of cervical cancer.Methods:MTT assay was used to analyze cells proliferation rate after treatmentwith different concentration of sorafenib for24h、48h、72h, and analyze inhibition ofsorafenib combined with cisplatin, paclitaxel on the proliferation of Hela cells. Cellapoptosis was determined by Hoechst33258fluorescence; Immunofluorescence andWestern blotting was used to observe the expression of VEGFR-3and PDGFR-βinHela cells before and after treatment of sorafenib, and analyze the correlation betweenthe expression of VEGFR-3/PDGFR-βand the proliferation rate of Hela cells,cellsapoptosis index.Results:Sorafenib inhibited the human cervical cancer Hela cells growth in aconcentration-dependent manner, with the increase of drug concentration, cellproliferation rate decreased.(P<0.05). When different concentrations of sorafenibcombined with cisplatin and paclitaxel, the cell proliferation rate decreased with theincreasing concentration of sorafenib, and the inhibition rate were higher thancisplatin、paclitaxel alone. In Hela cells, sorafenib showed the greatest synergisticeffect with the two cytotoxic drugs. Sorafenib can make Hela cells apoptosis. Theapoptotic cells, nuclear small, pyknosis, fluorescence enhanced obviously.we canobserved the expression of VEGFR-3and PDGFR-β in Hela cells, The greenfluorescence was reduced with the treatment of sorafenib, the expression of VEGFR-3and PDGFR-βin Hela cells were down, the expression of VEGFR-3and PDGFR-βwas positively correlated with the proliferation of Hela cells, and negative correlation with the apoptosis Hela cells.Conclusions:1:Sorafenib can inhibit the proliferation of Hela cells.2: Sorafenibcombined with cisplatin, paclitaxel inhibit the proliferation of Hela cells,and theinhibition is synergistic effect.3:Sorafenib inhibit Hela cells proliferation probably byinhibiting the expression of VEGFR-3and PDGFR-β in Hela cells.
