ObjectiveTo explore the microwave heating cisplatin-ferric powder reduce suspensioninduced apoptosis in pig’s liver and the relationship between TNF-α、Bcl-2and Baxprotein expression.MethodsThe pigs were randomly divided into the control group and the microwavetreatment group, The microwave treatment group with needle injected cisplatin-ferricpowder reduce suspension to the formation of hollow inside after ablation ofPig’s liver,Local heating with microwave apparatus for medical,the control groupwere not processing. Using TdT-mediated dUTP Nick-End Labeling (TUNEL)technical analysis to detect the apoptosis, Immunohistochemistry was used to detectthe protein expression levels of apoptotic cell-associated gene TNF-α、Bcl-2andBax.Results1、The microwave treatment group and the control group within the liverapoptotic cells were visible,the average apoptosis index(%) in the microwavetreatment group was7.55±1.74,and the apoptosis index(%) was5.33±1.45in thecontrols;Comparing the two groups, the microwave treatment group within the liveraverage cell apoptosis index (AI%) is significantly higher than that of the controls,the difference was statistically significant (P <0.01).2、TNF-α protein average positive expression rate(%) in the microwave treatmentgroup was11.70±3.02,the TNF-α protein average positive expression rate(%) was8.20±1.43in the controls;Comparing the two groups,the average positiveexpression rate of TNF-α protein in the microwave treatment group was obviouslyhigher than that of the controls,the difference was statistically significant (P <0.01). 3、Bcl-2protein average positive expression rate(%) in microwave treatmentgroup was6.61±1.74,the Bcl-2protein average positive expression rate(%) was9.81±3.01in the controls;Comparing the two groups, the average positive expressionrate of Bcl-2protein in the microwave treatment group was obviously lower thanthat of the controls,the difference was statistically significant (P <0.05).4、Bax protein average positive expression rate(%) in the microwave treatmentgroup was13.37±3.40,the Bax protein average positive expression rate(%)was9.81±2.80in the controls;Comparing the two groups,the average positive expressionrate of Bax protein in the microwave treatment group was obviously higher thanthat of the controls,the difference was statistically significant (P <0.05).5、The average ratio of Bax/Bcl–2in the microwave treatment group was5.34±2.55,the average ratio of Bax/Bcl–2was3.29±1.59in the controls; Comparingthe two groups, the ratio of Bax/Bcl–2in the microwave treatment group wasobviously higher than that of the controls,The difference was statistically significant(P <0.05).Conclusions1, microwave heating cisplatin-ferric powder reduce suspension can effectivelyinduce the apoptosis in pig’s liver.2microwave heating cisplatin-ferric powder reduce suspension can effectivelyinduce the apoptosis in pig’s liver and the mechanism may be through the followingseveral ways to achieve:1) increased the expression of TNF-α protein;2) inhibited the Bcl-2protein expression;3) increased the expression of Bax protein;4) increased the ratio of Bax/Bcl–2.
