Objective:The study was designed to investigate the role of the SIRT1mRNA expression in thedevelopment of coronary heart disease as well as in the glucose and lipid metabolism. Through thedrug intervention,we do the preliminary research and discunssion of left-hand amlodipine involvedin the mechanism of coronary heart disease.Methods:Stage11.Object42hospitalized patients from August2012to January2013who were diagnosed withcoronary artery disease by coronary artery angiography (the CAG),21cases of which was named“Amlodipine Group”,and the athers was named “Foundation treatment Group”.During the sameperiod,we selected anather21patients whose CAG results were negative,we named them “ControlGroup”.2. The Experimental MethodsWe separated peripheral blood mononuclear cells(PBMC) of the objects,extracted total RNAand through the reverse transcription,quantitative PCR,we detected the human PBMC SIRT1mRNA expresion level. At the same time,we detected the fasting blood glucose and lipid of allpatients. At last,we analysed the relationship between SIRT1mRNA and fasting blood glucose andlipid so called “Antherosclerosis Related Facters” among3groups.Stage21.Object“Amlodipine Group”(21cases)and “Foundation treatment Group”(21cases)were selected.“Amlodipine Group”(21cases) taked orally amlodipine besylate tablets5mg/day for6months.2. The Experimental MethodsWe separated peripheral blood mononuclear cells(PBMC),extracted total RNA and throughthe reverse transcription,quantitative PCR, detected the human PBMC SIRT1mRNA expresionlevel. We detected the fasting blood glucose and lipid of both two groups as well. At last,weanalysed the relationship between SIRT1mRNA with fasting blood glucose and lipid,and thechange of them before treatment and after. Result:1.In this experiment,before taking drug treatment,SIRT1mRNA expression in AmlodipineGroup(p=0.007) and Foundation treatment Group(p=0.032)were decreased,but there was nodifference between Amlodipine Group and Foundation treatment Group (p=0.568).2.6months afterdrug intervention, SIRT1mRNA expression in Amlodipine Group was higher than Foundationtreatment Group (p=0.033).3.SIRT1mRNA level in Foundation treatment Group and AmlodipineGroup were not difference from6month ago.4.Pearson and Spearman Analysis showed that FBG(r=-0.359,p=0.004), TC(r=-0.252,p=0.047), TG(r=-0.311,p=0.013), LDL(r=-0.344,p=0.006)had negatve correlated with SIRT1mRNA, and HDL(r=0.335,p=0.007) had positive correlatedwith it.5.6months after drug intervention, Pearson Analysis showed that FBG(r=-0.396,p=0.037),TC(r=-0.379,p=0.047), TG(r=-0.407,p=0.032), LDL(r=-0.469,p=0.012)had negatve correlatedwith SIRT1mRNA, and HDL(r=0.382,p=0.045) had positive correlated with it.6.After drugintervention,FBG(p=0.031), TC(p=0.02),TG(p=0.009)were decreased in Foundation treatmentGroup,TC(p<0.001)was decreased in Amlodipine Group.7.There were not difference before andafter the drug treatment about both FBG and Lipid level.Conclusion:1.SIRT1mRNA express in human’s peripheral blood mononuclear cells(PBMC).2. SIRT1mRNA level were decreased in both Amlodipine Group and Foundation treatment Groupbefore drug treatment which indicated that SIRT1mRNA probably participated in the formationand development of the coronary heart disease(CHD).3. SIRT1mRNA level in Amlodipine Group was higher than that in Foundation treatmentGroup,which show that amlodipine may be participated in the expression of SIRT1mRNA and thebiological effects of it.4.There were negative correlation between SIRT1mRNA with FBG, TC, TG and LDL–c andpositive correlation between SIRT1mRNA and HDL-c before drug treatment,as well as after thedrug intervention.
