Objective:To study the association between pigment epithelial dervied factor(PEDF)Gene polymorphisms and diabetic retinopathy in Chinese Han population Fujianregion.Meanwhile,analyzing the risk factors for2diabetic retinopathy(DR),to offer anew way for preventing and curing DR.Methods:Two hundreds and fifty six cases of T2DM who were diagnosed in the Firstaffiliated Hospital of Fujian Medical University from September,2011toAugust,2013,the gender,age,diabetes duration,height,weight,Body MassIndex(BMI),systolic blood pressure,diastolic blood pressure,glycatedhemoglobin(HbA1C) and total cholesterol(TC) were analyzed.The patients weredivided into two groups:Diabetic retinopathy (DR)group(n=128) and Non-diabeticretinopathy(NDR) group(n=128).Genomic DNA were extracted from peripheral whiteblood cell.The polymorphism of the two single nucleotide polymorphisms(SNPs) ofPEDF gene: rs12150053and rs58697961were determined by Matrix assisted laserdesorption ionization time of flight mass spectrometry(MALDI-TOF MS).Thefrequencies distrubition of genotype and allele between DR group and NDR groupwere compared by chi square (P <0.05),and the risk factors of type2DR wereanalyzed by logistic regression applying SPSS18.0statistical software.Results:(1)Results of Independent samples t test showed that the difference of diabetesduration and HbA1C between DR group and NDR group was statisticalsignificance(P<0.001).Results of univariate logistic regression analysis showed thatthe difference between diabetes duration and DR was statistical significance.With thedevelopment of diabetes duration,the risk rate of DR increased,its OR(95%CI) were1.812(1.268-2.590). The difference between HbA1C and DR was statisticalsignificance.With the increasing of HbA1C,the risk rate of DR increased,its OR(95%CI) were3.005(1.926-4.690). (2)The distribution of allele and genotype of rs12150053and rs58697961wasaccordance with Hardy-Weinberg’s Law of Equilibrium,and exhibits therepresentative of population(P>0.05).(3)The genotype frequencies of rs12150053of PEDF gene in DR group were9.4%,52.1%and37.5%and25.8%,50.8%and23.4%in NDR group for TT,TC andCCgenotypes respectively.The difference of the genotype frequencies between thetwo groups was statistical significance(P=0.001).The difference of allele frequenciesof rs12150053of PEDF gene between the two groups was statisticalsignificance(P=0.001).Results of univariate logistic regression analysis showed thatthe difference between DR and PEDF genotype was statistical significance,its OR(95%CI) was1.978(1.352-2.894),and still statistically significant after adjusting fordiabetes duration and HbA1C.Type2diabetic patients with TC/CC genotype hadhigher risk of DR,3.358(1.644-6.859) times than the one with CC genotype,and stillstatistically significant after adjusting for diabetes duration and HbA1C.Type2diabetic patients with C allele had higher risk of DR,1.868(1.311-2.661) times thanthe other.(4)The genotype frequencies of rs58697961of PEDF gene in DR group were14.8%,55.5%and29.7%and25.0%,52.3%and22.7%in NDR group for GG,GA andAA genotypes respectively.The difference of the genotype frequencies between thetwo groups was no statistical significance(P=0.097).The difference of allelefrequencies of rs58697961of PEDF gene between the two groups was no statisticalsignificance(P=0.051).(5)Three factors related to diabetic retinopathy were introducted into multivariatenon-conditional logistic regression equation,which included diabetes duration, HbA1Cand rs12150053of PEDF gene TC/CC genetype.The ORs(95%CI) wereas follow:1.072(1.009~1.138),1.121(1.007~1.247)and3.518(1.684~7.346).Conclusions:The difference of diabetes duration,HbA1C and rs12150053of PEDF gene TC/CCgenetype between DR group and NDR group was statistical significance.The one with the longer diabetes duration, the increasing of HbA1C and rs12150053of PEDF geneTC/CC genetype were related to high risk of diabetic retinopathy.
