Objective To observe the effects of Bone marrow-derived mesenchymal stem cells(BMSC)on glomerular podocyte injured by lipopolysaccharide (LPS) and theexpression of some related protein, exploring the possible protection mechanism ofBMSC to injured podocyte.Methods Isolating and cultivating BMSC from BALB/C mice femur by the wholebone marrow culture method. Appraisaling the podocytes by immunocytochemistrystaining.Experiment is divided into four groups: Control group, BMSC group, LPSgroup and LPS+BMSC group. After24hours, observing each experimental grouppodocyte form under inverted phase contrast microscope;detecting the expression ofthe mRNA and protein of Nephrin, CD2AP, Synaptopodin, TRPC6by the methods ofRT-PCR and Western-blot.Results1. FACS display that the positive expression rate of3rd generation mouseBMSC surface antigen CD44is98.7%±0.9%, CD90is98.1%±1.4%, CD29is96.1%±2.4%, while the positive expression rate of CD11b/c is14.7%±0.6%,CD34is1.98%±0.5%.2. By immunocytochemistry, undifferentiated podocytes wasoval and have small nuclei and there was no brown substance sesi in cytoplasm, didnot express podocyte-specific cytoskeletal proteins Synaptopodin;In contrast, nucleiof podocyte differentiation and maturation changed into a circle and the cytoplasmseen a lot of brown Silk-like, expressing Synaptopodin.3. cell morphology wasobserved in all experimental groups:LPS group podocyte cell shrinkage, losing theirdistinct appearance and the gravel-like material Increased intracellular;LPS+MSCgroup cells were still visible of the original cell morphology, cell shrinkage was notobvious and intracellular gravel material decreased production.4. the expression ofNephrin, CD2AP, Synaptopodin in LPS+MSC group were up-regulated than LPSgroup (P<0.05) while TRPC6was down-regulated (P<0.05).Conclusion1. BMSC can inhibit LPS-induced podocyte injury.2. BMSC protective effect on podocytes may be associated the expression of Nephrin、CD2AP、Synaptopodin、TRPC6expression.
