Objective: we observe the effect of curcumin on redox and insulin resistancestate of myocardial tissue. We also determine the expression of GLUT1, GLUT4,TXNIP, p-AMPK, AMPK, Nrf2, NQO-1in myocardial.Method: Twenty C57BL/6J mile male mice were divided into four groupsrandomly. we randomly chose two groups and gave them intraperitoneal injection ofstreptozocin (STZ) to making the diabetes model. we feed curcumin (10mg/kg,1kgof high-fat diet containing10mg of curcumin) to one of the group, and give the othergroup high fat feed.For the other two groups, we gave One group low fat feed, andthe other one high fat feed. The mice were killed at the intervention of15th day. Weused MDA kits to determine the redox of myocardial and ELISA kits to examine theserum insulin. Western blot was applied to determine the expression of GLUT1,GLUT4, TXNIP, p-AMPK, AMPK, Nrf2, NQO-1in myocardial.Results:(1) Short-term curcumin intervention can improve the sugar metabolicdisorder in diabetic mice which were induced by STZ(on the fifth day group ofSTZ+HFD22.1±2.2vs the group of STZ+HFD+Cur17.8±1.8,P<0.01;on the tenthday the group of STZ+HFD25.4±3.2vs the group of STZ+HFD+Cur18.4±1.4,P<0.01)(2) Curcumin can improved the level of insulin in diabetes mice (the groupof STZ+HFD4.3±1.0vs the group of STZ+HFD+Cur12.1±4.0,P<0.05).(3)Themyocardial MDA was significantly decreased compared with the STZ group.(thegroup of STZ+HFD2.3±0.5vs he group of STZ+HFD+Cur组1.3±0.3,P<0.01).(4)The glucose uptake capacity of myocardial tissue results showed that,the curcuminintervention group’s TXNIP expression were reduced, while the glucose transportersGLUT4and GLUT1increased significantly and the p-AMPK. Conclusions:(1)Weobserved that in mice model of DM, curcumin can obviously decrease blood glucoseand increase serum insulin levels, thus to reduced the level of blood glucose.(2)Curcumin can obviously decrease the MDA content in myocardial, thus relieving theoxidative stress injury; The mechanism may be that curcumin can upregulated the levels of NQO-1, and phosphorylation of AMPK, reduced the level of TXNIP protein.(3)Curcumin can improve the ability to absorb glucose of the myocardial of diabeticmice By the way of down-regulating TXNIP expression and up-regulating GLUT1and GLUT4expression. Short-term curcumin intervention had no obvious effect onthe Nrf2antioxidant system.
