Objective: To investigate the changes of orexin and orexin receptor-1(OX1R)expression in prefrontal cortex of coma animal models after traumatic brain injurywith median nerve electrical stimulation, preliminarily exploring the effect andpossible mechanisms of median nerve electrical stimulation in come and wake.Methods: SD rats were divided into3groups: control group, sham-stimulationgroup, stimulation group. Rat model of persistent coma state after traumatic braininjury was established.The prefrontal cortex was extracted from rat brain of eachgroup at6,12,24h after median nerve electrical stimulation respectively; ELISA,immunohistochemical, Western-blot techniques were used to detect the expressions oforexin and OX1R.Results: The results of ELISA showed that Orexin-A of prefrontal cortical instimulation group was higher than the control group and the sham-stimulationgroup,with the difference being statistically significant(P﹤0.05).The expression ofOrexin-A showed an increasing trend in each group,and there were significantdifferences in the level of the Orexin-A between the stimulation group and the othertwo groups(P﹤0.05)at6,12,24h respectively.The results of Western-blot indicatedthat the level of OX1R was higher than the other two groups,and the difference wasstatistically significant(P﹤0.05). The expression of OX1R also showed an increasingtrend in each group, there were significant differences in the level of the OX1Rbetween the stimulation group and sham-stimulation group(P﹤0.05)at6,12hrespectively, but there were not significant differences in the level of the OX1Rbetween the stimulation group and sham-stimulation group(P﹤0.05)at24h. Theresults of IHC indicated that the level of OX1R was higher than the other two groups,and the difference was statistically significant(P﹤0.05). The expression of OX1Ralso showed an increasing trend in different time of three groups,and the differencewas statistically significant(P﹤0.05). There were not significant differences in thelevel of the OX1R between the stimulation group and sham-stimulation group(P﹤0.05)at6h,but the difference was statistically significant(P﹤0.05)at12h,24h respectively.Conclusion:1.The expression of Orexin-A and OX1R in prefrontal cortex ontraumatic brain injury induced coma for rats by median nerve electrical wasup-regulated,which is related to the effect of awaking-promoted of MNS in comaratsafter traumatic brain injury.2. The Orexin-A and OX1R protein expression level in prefrontal cortex ontraumatic brain injury induced coma for rats was increased at6h,peaked at24h.whichis related to the effect of physiological emergency reaction, in order to protect thedamaged neurons and reduce the damage of neurons.
