Objective:Nowadays lung cancer has become the malignancy which has the quickestincrease in morbidity all over the world. Lacking of an efficient way to have a earlydiagnosis, people urgently expect to get rid of lung cancer by having a healthy diet.Chemorevention is a key factor to prevent cancer. A large amount of experiments andstudy on epidemiology proves that healthy diet and lifestyle can reduce the risk ofcancer to a large extent. Tea is one of the most popular drinks in the world. Green teahelps to avoid cancer, because of that, people expect to get away from lung cancer bydrinking green tea in our life. Meanwhile, building pulmonary malignant tumoranimal models is a necessary way to assess the curative effect of preventing lungcancer. This topic through the benzopyrene, this caused by smoking and air pollution,in People’s Daily life most frequent contact with highly active carcinogen, through thelung puncture operation, the test carcinogenic injection in rats, to build the ratpulmonary malignant tumor animal models, through this model, to evaluated theintervention effects of green tea on pulmonary malignant tumor, And on this basis, thefurther research of green tea on rat pulmonary malignant tumor on the p53proteinand survivin protein expression. At the same time, by enzyme-linked immuno sorbentassay, to detect the expression of VEGF in rat animal model, preliminary proven togreen tea on the intervention mechanism of pulmonary malignant tumor.Methods:Building model of rat lung malignancy. Using3,4-benzopyrene powder mixedin corn oil solution, mixture mixed solution, every time all fixed injection contains3,4-benzopyrene2mg dose of corn oil mixture of0.2ml. In use after intraperitonealinjection of10%chloral hydrate anesthesia in rats between3-4ribs on the right sidepercutaneous lung biopsy. Control group using pure corn oil solution lung punctureinjection. Each group every two weeks for one times, a total of four times.In green tea intervention pulmonary malignant tumor in the experimental group,using only the only drinks green tea as a daily chemical prevention. While drinking group and the control group using water as a daily drinks. Observation rat pulmonarymalignant tumor, disease processes by immunohistochemical techniques methods ongreen tea and water group in the rat lung tumor p53protein and survivin proteinexpression difference. At the same time, by enzyme-linked immuno sorbent assay todetect differences between groups in the rat serum VEGF expression.Results:All pure accept corn oil solution lung puncture injection control group rats, notumor found in52weeks test cycle. Group of drinking water in52weeks malignanttumor incidence was65%, green tea set of incidence of malignant tumors in52weeksdecreased obviously, only20%. Immunohistochemistry results showed that green teaset of animal model of pulmonary malignant tumor tissue gene expression level,cutting survivin protein and raising p53protein expression, compared with watergroup, on the expression differences statistically significant (P<0.05). Enzyme-linkedimmuno sorbent assay results showed that green tea set of VEGF expression, andcompared with the group of drinking water, on the expression differences statisticallysignificant (P<0.05).Conclusion:Green tea for3,4-benzopyrene pulmonary malignant tumor caused by acarcinogenic agent has evident in the prevention and intervention effect. By raisingthe expression of p53protein, cut survivin protein and the expression of VEGF is theimportant mechanism of the green tea to prevent lung cancer.
