BackgroundBreast cancer is the most common malignant disease during women worldwidewhich is the main reason of dying to malignant in women. The incidence of Europeand the United States is highest in the whole world.The breast cancer incidence andmortality rates among Chinese women were increasing rapidly, especially in ruralarea during the recent ten years, though they were still in low level worldwidely.Exact statistics at present still has no national breast cancer incidence in ourcountry,according to recent statistics of Shanghai,the city’S breast cancer incidencehas rapidly risen from the seventh female cancer to the first during ten years.Different types and malignant degree of breast cancer have a great differenceresponse to the chemotherapy and prognosis. Therefore, in order to forecast themalignant degree of tumor better, may also provide a promising target for thetreatment of breast cancer.4’phosphatidylinositol kinase Ⅱα (PI4KⅡα) is widelyexpressed in human tissues, especially enriched in brain and kidney. PI4KⅡα ismembrane protein,which localise to the trans-Golgi network (TGN), endosomes andplasma membrane (PM). The mammalian alpha and beta PtdIns4KIIs are bothmembrane proteins of~55kDa. Researchs have shown that, overexpression ofPI4KⅡα in tumor cells, which promotes tumor growth through of angiogenesis inthe regulation of tumor growth. Blocking PI4KIIa expression in tumor cellssuppresses tumor growth in vivo. Thus, we can conclude that PI4KIIa has animportant role in tumor progression.Through this research on the expression level of PI4KⅡα in invasive breastcancer and normal tissue mRNA, investigate whether the high expression in breastcancer. It analyses the expression level of PI4KⅡα whether it is correlation withclinical indexes and pathological indexes,such as tumor size,ER, PR, Her-2andlymph node metastasis. Materials and Method1.MaterialI randomly selected48cases of breast cancer tissue after breast surgeryoperation in the First Affiliated Hospital of Nanchang University During the period of2010to2011.Meanwhile,48cases of normal breast tissue are from more than5cm ofthe cancer tissue edge. These tissues were preserved in refrigerator of negative80centigrade. Patients’ medical records which contain pathological indexes havecollected to the database.2. Method(1)Reverse transcription polymerase chain reaction (RT-PCR):Extraction of total RNA,reverse transcription into cDNA,select the Actin as areference,1%agarose gel electrophoresis and scanning with ultraviolet.(2)Real time quantitative PCR(Q-PCR):The objective of using Q-PCR is to calculate relative expression quantity ofPI4KⅡα in the cancer tissue (T) and normal tissue (N),and calculate their ratio(T/N).(3)Statistical analysis:All the data are analyzed with SPSS17.0software. Rank sum test can be appliedto see if PI4K Ⅱα expression with tumor size, ER, PR, Her-2and lymph nodemetastasis have the relation. P<0.05is considered statistically significant.Results(1)After the experiment,during48cases of breast cancer tissue and normaltissue, only39cases are for the effective data.(2)The study show that the relative expression amount of PI4K Ⅱα in invasivebreast cancer tissue and normal tissue from the same patient,which there are5casesof overexpression,accounting for12.8%;23cases of low expression,accounting for58.9%.(3)Between PI4KⅡα and other clinical and pathological indexes: PI4KⅡαhas a correlation with tumor size that is considered statistically significant(P<0.05).But ER, PR, Her-2and lymph node metastasis are not consideredstatistically significant. Conclusion(1) The expression level of PI4KⅡα in invasive breast cancer of mRNA,comparing to the normal tissue of the same patient, have no specific high expression.(2) There is significant correlation between PI4KⅡα and tumor size, P<0.05,is considered statistically significant.Researchs showed that the expression ofPI4KⅡα may upregulate as the tumor growing up, or PI4KⅡα could regulate tumorgrowth.But ER, PR, Her-2and lymph node metastasis are not considered statisticallysignificant.
