Pemetrexed Inhibitory Effect On Human Colon Cancer HT29Cells And Its Effect On The Expression P21^WAFI

Backdrop and objective：The incidence of colon cancer in various regions of our country’s is high.Recentstudies have shown a close contaction between P21WAFIand the development、invasion and metastasis of colon cancer.In the meantime the research betweenpemetrexed and P21WAFIalso has come out.The goal of this study is to detect P21WAFIexpression in different drug acting time and concentrationn,and to find the connectionbetween the change of P21WAFIexpression and HT29proliferation and apoptosis.Methods：The expressions of P21WAFIin HT29is detected by RT-PCR, Western blot afterdifferent drug concentrations and acting time of pemetrexed and5-FU. In addition,the best reaction time and concentration are selected.The two drugs effects on the biological activity of HT29in optimalconcentration and reaction time are compared by MTT assay, flow cytometry.Results：1、 RT-PCR, Western blot showed that pemetrexed and5-FU optimalconcentration and reaction time are1000μmol/L,48h.2、P21WAFIof two drugs under optimal conditions are0.93±0.19,0.85±0.15, P<0.05in the RT-PCR; the Western blot results are1.59±0.13,1.33±0.14, P <0.05.3、The OD values are0.14±0.07,0.21±0.06, P <0.05in MTT assay, they aresignificantly lower than the control group0.51±0.11.The rate of apoptosis are37.03%，14.02%, P <0.05, significantly higher than the control group5.03%in flowcytometry.Conclusion：1、P21WAFIupregulation maybe is associated with the inhibition of proliferationand the promotion of apoptosis in HT29cells after pemetrexed and5-FU.2、Pemetrexe has a strongger effect on inhibition proliferation and promotionapoptosis in HT29cells compared with5-FU at the same time the drug concentration.
